In a sealed tube, a mixture of Step 1SM-4 SM-5a B-1a2-Chloro-3-fluoro-6-methylpyrimidine SM-4 (1.0 g, 6.9 mmol), 1-Boc-4-(methylamino)- piperidine SM-5a (2.95 g, 13.7 mmol), Pd2(dba)3 (190 mg, 0.2 mmol), DavePhos (324 mg, 0.8 mmol) and LiHMDS (34.35 mL, 134.35 mmol) are taken-up in dry THF (20 mL) and the resulting mixture is flushed with argon and stirred for 45 min at 80 C. The reaction mixture is diluted with DCM, washed with H20 and brine. The organic layer is dried, filtered and concentrated under reduced pressure. Isolute is added, the solvent is removed in vacuo and the residue is purified via NP-MPLC.
