Acid green 5 is an organic sodium salt having 3-[(ethyl{4-[(4-{ethyl[(3-sulfonatophenyl)methyl]amino}phenyl)(4-sulfonatophenyl)methylidene]cyclohexa-2,5-dien-1-ylidene}azaniumyl)methyl]benzene-1-sulfonate as the counterion. It is the standard dye in North American for staining collagen and is also used extensively in plant histology. It has a role as a histological dye. It contains an acid green 5(2-).|Light green SF yellowish is a green triarylmethane dye that is used in the preparation of the staining solution which is widely used as a counterstain. It is used in histological applications and other labratory settings, and usually exists as a disodium salt. The maximum absorption of light green SF yellowish is at 630 (422) nm. Although it has been used as a food colorant, it was discontinued from the market due to low popularity which are possibly due to low durability of the dye and increased tendency of the dye to fade. Light green SF yellowish is also available as sterile strips used as a diagnostic agent when superficial corneal or conjunctival tissue change is suspected.

FD&C Green No. 2's production and use as a dye(1) may result in its release to the environment through various waste streams(SRC).
TERRESTRIAL FATE: Acid dyes, such as FD&C Green No. 2, typically exhibit very low solubilities in octanol and have water solubilities that exceed 100 g/L. Therefore, these compounds would be expected to remain in the water column in the aquatic environment and show little affinity for organic matter(1) which suggests a potential to leach in soil(SRC). Volatilization of FD&C Green No. 2 from moist soil surfaces is not expected to be an important fate process(SRC) given an estimated Henry's Law constant of 1.4X10-36 atm-cu m/mole(SRC), using a fragment constant estimation method(2). FD&C Green No. 2 is not expected to volatilize from dry soil surfaces(SRC) based upon an estimated vapor pressure of 2.97X10-42 mm Hg at 25 °C(SRC), determined from a fragment constant method(2).|AQUATIC FATE: Acid dyes, such as FD&C Green No. 2, typically exhibit very low solubilities in octanol and have water solubilities that exceed 100 g/L. NIOSH (NOES Survey 1981-1983) has statistically estimated that 18,350 workers (7,859 of these were female) were potentially exposed to FD&C Green No. 2 sodium in the US(1). Occupational exposure to FD&C Green No. 2 may occur through dermal contact with this compound at workplaces where FD&C Green No. 2 is produced or used(SRC).
Drug Information
Light green SF exhibits high plasma binding.|Studies in rats and dog involving oral administration of light green SF indicate that only small amounts of the dye (<5%) are absorbed and excreted, mainly in the feces. Following its iv injection in rats, about 20% of the dye was excreted in the bile within four hours.
/SRP:/ Immediate first aid: Ensure that adequate decontamination has been carried out. If patient is not breathing, start artificial respiration, preferably with a demand valve resuscitator, bag-valve-mask device, or pocket mask, as trained. Perform CPR if necessary. Immediately flush contaminated eyes with gently flowing water. Do not induce vomiting. If vomiting occurs, lean patient forward or place on the left side (head-down position, if possible) to maintain an open airway and prevent aspiration. Keep patient quiet and maintain normal body temperature. Obtain medical attention. /Poisons A and B/|/SRP:/ Basic treatment: Establish a patent airway (oropharyngeal or nasopharyngeal airway, if needed). Suction if necessary. Watch for signs of respiratory insufficiency and assist ventilations if needed. Administer oxygen by nonrebreather mask at 10 to 15 L/min. Monitor for pulmonary edema and treat if necessary ... . Light Green SF is prepared commercially by condensing ethylbenzylaniline with benzaldehyde, followed by sulphonation and oxidation, and, finally, conversion to the sodium salt.

Light Green SF is not included in the list of colorants permitted for use in food in the European Economic Communities ...|Color additives were initially regulated in the United States under the U.S. Department of Agriculture's (USDA) Bureau of Chemistry. In 1906, the Food and Drugs Act was passed by Congress, which prohibited the use of poisonous or deleterious colors in confectionery and the coloring or staining of food to conceal damage or inferiority. In 1927, responsibility of the Food and Drugs Act was transferred to FDA.
