Extract 150g of acetone powder from the posterior leaves of pig pituitary gland, add 375ml of acetone, stir, add 2400ml of glacial acetic acid, heat in a water bath at 70°C for half an hour, cool to below 35°C, filter, wash the filter residue with 300ml each of glacial acetic acid and acetone, and combine Wash and filtrate to obtain extract. Pituitary posterior leaves of pig acetone powder [acetone; glacial acetic acid]→[70℃] extract separation and precipitation. Add 14ml saturated NaCl solution to the extract, add 1390ml acetone to precipitate the precipitate, refrigerate overnight, and filter the next day to obtain 4000ml filtrate. If it is not clarified, it can be filtered with No. 4 vertical melting funnel, and the filter residue can recover prolactin. Add the same volume of ether under stirring, let it stand for a while, absorb the supernatant, filter, and wash the precipitate with acetone to remove acetic acid, then wash twice with anhydrous ether, and dry with phosphorus pentoxide to obtain a crude product. Extraction solution [saturated NaCl, acetone] → filtrate [diethyl ether] → MSH crude refined product 22g dissolved in 620ml acetic acid (0.1mol/L), added 1 mol/L hydrochloric acid, water and acetic acid (0.1mol/L) washed Oxidized cellulose was 3.8g, stirred for 4h, filtered by No. 3 vertical melting funnel, and oxidized cellulose was recovered. Wash 3-4 times with the aforementioned acetic acid, wash several times with distilled water, place in a beaker after suction filtration, add 82ml acetic acid (concentration same as before) and stir for half an hour, after filtration, wash once with 82ml 0.1mol/L hydrochloric acid and combine the filtrate. Adjust the pH to 6.5 with ammonia water, collect the supernatant by centrifugation, add 1L of cold acetone to refrigerate, centrifuge, collect the precipitate and wash it twice with acetone, wash once with ether, and dry in a phosphorus pentoxide dryer to obtain a refined product. MSH crude product [acetic acid, oxidized cellulose, HCl] → [ammonia, acetone, pH6.5] finished product.
Suitable for skin and hair melanin deficiency. 5% eye drops can improve the eye~"s ability to adapt to darkness. Effective for retinal degeneration or pigment degeneration.
