D-isoleucine is the D-enantiomer of isoleucine. It has a role as a bacterial metabolite and a Saccharomyces cerevisiae metabolite. It is an isoleucine and a D-alpha-amino acid. It is a conjugate base of a D-isoleucinium. It is a conjugate acid of a D-isoleucinate. It is an enantiomer of a L-isoleucine.

Using L-isoisoleucine as raw material, after acylation reaction with acetic anhydride, the product is treated with acylating enzyme to separate L-isoisoleucine and then hydrolyzed.
