Z-GLU(OME)-OH Use and Manufacturing

N-CBZ-glutamic acid 5-methyl ester (25 g, 0.1 mol) was dissolved in t- butyl acetate (250ML) and 70% perchloric acid (1.5 ML) was added. The solution was stoppered, stirred and left to stand at room temperature for 48 hours. The flask was opened carefully and added dropwise to a saturated solution of NAHCO3. When the addition was complete, the mixture was extracted with ethyl acetate and the organic phase washed with water and brine, dried over magnesium sulfate and evaporated to dryness. The oily product was purified by column chromatography over silica gel eluted with hexane ethyl acetate (4: 1).
