Polyoxin B Use and Manufacturing
Technological process of biosynthesis of polyoxin: sand tube strains → bevel strains [10~14d, 26℃] → shake flask seeds [36~48h, 28℃] → seed pot seeds [24h, 28℃] → fermentation [90~120h, 28℃] Put in the tank, acidify the fermentation broth (pH value 3~4)→concentrate→dry→packaging strain Streptomyces aureo chromogenes. The formula of slant culture medium for slant culture is as follows. Maltose 0.5%; corn flour 0.5%; yeast extract 0.4%; agar 2%; pH value 6.3, cultured at 26℃ for 10-14 days after inoculation. Shake flask seed culture medium formula: soybean cake powder (cold pressed) 5%; soybean oil 2%; glucose 1%; KH2PO4 0.1% adjust pH value 6.5 and add calcium carbonate 0.3%. The tank pressure was 4.9×10Pa, the tank temperature was 28°C, the ventilation rate was 1:1, and the mechanical stirring was used for 24 hours. Fermentation medium formula: fermented soybean cake powder 2%; corn meal 1.5%; caramel 4%; yeast powder 0.4%; fish meal 0.5%; sodium chloride 0.1%; antifoam oil 0.5%, adjust the pH value 6.5 and add calcium carbonate 0.3%. The tank pressure is 0.05MPa, the tank temperature is 28℃, the ventilation rate is 1:1 (less before and after, more during the middle), and the fermentation time is 90~120h. After being put into the tank, it is acidified by adding oxalic acid, and the pH value is 3-4. Concentrated fermentation broth After the fermentation broth was acidified, it was concentrated under reduced pressure with a vacuum film concentrator. The liquid temperature is 40~60℃, the pressure is 1.33×10-4~1.47×10-4Pa, and the flow rate is 250kg/h. The dried and packaged concentrated liquid is sprayed into the drying tower for spray drying. The temperature of the hot air into the drying tower is 150℃, the exhaust temperature is 75℃, the negative pressure in the tower, the original powder is crushed, and the titer is adjusted to 1.5×10 units (1.5%) with talc powder.
