Pfizer's ADC Portfolio:

Pfizer's foray into the ADC field began with the approval of its first ADC drug, Mylotarg (gemtuzumab ozogamicin), by the FDA in 2000. Initially, Mylotarg faced setbacks due to concerns over its clinical benefits and higher toxicity compared to chemotherapy. However, recognizing the urgent need for effective treatments in acute myeloid leukemia (AML), Pfizer successfully regained FDA approval for Mylotarg in 2017 for adult and pediatric patients with CD33-positive AML.

 

Besponsa (inotuzumab ozogamicin), Pfizer's second ADC drug, received marketing authorization in the European Union and the United States in 2017 for the treatment of relapsed or refractory precursor B-cell acute lymphoblastic leukemia (ALL). Besponsa, a third-generation ADC targeting CD22, demonstrated moderate market performance, with revenues reaching $236 million in 2023, reflecting a 7.8% year-on-year growth.

 

Pfizer's Strategic Acquisitions:

Pfizer's commitment to ADCs became even more evident with its significant acquisition of Seagen for a staggering $43 billion in 2023, setting a new record for ADC-related transactions. Prior to the acquisition, Pfizer already had two commercialized ADC products, Mylotarg and Besponsa, which were acquired through its $68 billion purchase of Wyeth in 2009. The Seagen acquisition further boosted Pfizer's ADC portfolio by adding four approved ADC products: Adcetris, Padcev, Tivdak, and Disitamab vedotin, developed by its partner Zymeworks. Additionally, Seagen brought along numerous ADC candidates in various stages of clinical development.

With the completion of the Seagen acquisition, Pfizer now boasts a total of six approved ADC drugs, solidifying its position as the company with the highest number of ADC products on the market. This achievement has positioned Pfizer as an industry leader, shaping the future of ADC therapy and underscoring its commitment to innovative cancer treatments.
