To improve delivery efficiency and minimize therapeutic side effects, researchers have developed a variety of delivery vehicles to encapsulate mRNA, using natural or synthetic materials (e.g. lipids, polymers, peptides, antibodies, small molecules or metals) to make nanoparticles with different geometries (nanoparticles, particles, conjugates, solid devices or hydrogels, etc.) to safely deliver mRNA to target cells to express normal proteins.

CPPs, discovered in 1988, are short sequence peptides that help cells to take up a variety of molecules. Although their transmembrane mechanism is still unclear, CPPs have been successfully synthesized artificially and have great potential in drug delivery, protein, plasmid DNA and siRNA by virtue of their low charge density and excellent ability to cross cell membranes, and are mostly in the research exploration stage in the field of mRNA delivery. Another disruptive achievement in protein carriers is the SEND delivery technology developed in August 2021 by gene editing pioneer Feng Zhang and team. at the heart of SEND is the mammalian retrovirus-like protein PEG10, which has the potential to transport RNA between cells.

The researchers discovered that PEG10 is specifically designed to recognize and package mRNA gene sequences, and when the signal sequence is added to both ends of the RNA molecule, PEG10 is able to load the "cargo" RNA. The fusion technique was then used to add the viral fusion protein Fusogen to PEG10, allowing PEG10 to fuse with target-specific cell surfaces and deliver the cargo RNA to the cytoplasm. The study, published in Science, is still in the mouse stage, but I venture to guess that clinical trials will not be long in coming, as the mechanism borrows from naturally occurring RNA-carrying proteins and its safety and efficacy have long been tested by nature's evolution.

Currently, there are many innovative companies around the world investing in mRNA vaccine research and development, and the mRNA vaccine delivery technology in the marketed or clinical stage is still mainly LNP. The global TOP3 mRNA vaccine leaders are Moderna, BioNTech and CureVac, with multiple R&D pipelines.
