Appearance

This product is a white almond-shaped film-coated tablet with "GILEAD" engraved on one side and "4331" engraved on the bottom. It appears white after removing the coating.

Usage and Dosage

Recommended dose for adults and children aged 12 years and over (35 kg or more): For the treatment of HIV-1 or chronic hepatitis B: 300 mg (one tablet) once a day, orally, on an empty stomach or with food. The best course of treatment for chronic hepatitis B has not yet been determined. The safety and efficacy of this drug in children with chronic hepatitis B weighing less than 35 kg have not been studied. [See instructions for details]

Adverse Reactions

In both studies, nearly 1/3 of the patients in the FDA group experienced adverse reactions, but the incidence was similar to that in the control group. Common adverse reactions of FDA include: mild to moderate gastrointestinal discomfort (commonly diarrhea, nausea, vomiting and flatulence); hypophosphataseemia of the metabolic system (1% incidence), mild to moderate decrease in serum phosphate (12% in the FDA group at 24 weeks, 7% in the control group, and 15% in the FDA group at 58 weeks). There is a risk of lactic acidosis.

Precautions

This drug is contraindicated in patients with a prior allergy to any of the ingredients in this drug.

Special Population Medication

Precautions for children: Not yet clear. Precautions for pregnancy and lactation: Not yet clear. Precautions for the elderly: Clinical trials of tenofovir disoproxil fumarate did not include enough subjects aged 65 years or older to determine whether their responses differed from those of younger subjects. In general, elderly patients should be cautious in choosing doses, keeping in mind that they have decreased liver, kidney, and heart function, and are at increased risk of concurrent diseases or being treated with other medications.

Drug Interactions

1. This product can interfere with the blood concentration of didanosine. Tenofovir should be taken 2 hours before or 1 hour after didanosine. 2. The combined use of didanosine and this product can increase the blood concentration of didanosine, increasing the risk of pancreatitis. 3. This product is excreted from the kidneys through glomerular filtration and tubular secretion. When used in combination with other drugs that are secreted and excreted through the tubular secretion, it can increase each other's plasma drug concentration. Drugs that reduce renal function can also increase the blood concentration of this product. 4. This product can be used in combination with lamivudine, indinavir, lopinavir, and ritonavir to reduce the blood concentration of these drugs.
