Thioguanine Tablets

It is a commonly used purine metabolism antagonist drug that inhibits the purine synthesis pathway. It is a cell cycle-specific drug that is most sensitive to cells in the S phase. In addition to inhibiting the synthesis of cell DNA, it also has a mild inhibitory effect on the synthesis of RNA. This product is an analog of guanine. It must be converted into 6-TG ribonucleotides by phosphoribosyltransferase in the human body to be active. The action link of this product is similar to that of mercaptopurine. In addition, 6-TG ribonucleotides can prevent the phosphorylation of guanosine monophosphate (GMP) to guanosine diphosphate (GPD) by inhibiting guanylate kinase. After being metabolized to deoxyribose triphosphate, this product can be incorporated into DNA, thereby further inhibiting the biosynthesis of nucleic acids. Mercaptopurine does not have this effect. This product has effective cross-resistance with mercaptopurine, and can improve the efficacy when used in combination with drugs such as cytarabine.

This product is white tablets.

1. The induction remission period and continuing treatment period of acute lymphocytic leukemia and acute non-lymphocytic leukemia; 2. The chronic phase and blast crisis phase of chronic myeloid leukemia.

Common adult dosage, oral, initially 2mg/kg or 100mg/m2 daily, once a day or divided into several doses. If there is no clinical improvement and no leukocyte suppression after 4 weeks, the daily dose can be increased to 3mg/kg with caution. The maintenance dose is 2mg-3mg/kg or 100mg/m2 daily, once or divided into several doses. In combination chemotherapy, 75-200mg/m2 is taken once or divided into several doses for 5-7 consecutive days.

1. Common toxic reactions include bone marrow suppression, which may lead to leukopenia and thrombocytopenia; 2. Digestive system reactions: nausea, vomiting, loss of appetite and other gastrointestinal reactions and liver damage, which may be accompanied by jaundice; 3. Leukemia and lymphoma patients who begin treatment may develop hyperuricemia, and in severe cases, uric acid nephropathy may occur; 4. This product may inhibit testicular or ovarian function, causing amenorrhea or sperm deficiency, which is related to the dosage and course of the drug, and the reaction may be irreversible.

Patients with known hypersensitivity to this product are contraindicated.

Precautions for children: The usual dosage for children is 2.5 mg/kg per day, orally once a day or divided into two doses. Precautions for pregnancy and lactation: There is an increased risk of fetal death or congenital malformations. Avoid taking it within the first 3 months of pregnancy. Use with caution in lactating women. Precautions for the elderly: Elderly patients have poor tolerance to chemotherapy drugs, so supportive therapy should be strengthened during medication, and the condition and possible adverse reactions should be closely observed, and the dosage should be adjusted in a timely manner.

This product has the effect of increasing the blood uric acid content. Therefore, when used simultaneously with anti-gout drugs, the dosage of anti-gout drugs must be adjusted to control hyperuricemia and gout. When this product is used in combination with other anti-tumor drugs or radiotherapy that have bone marrow suppression, the effect of this product will be enhanced. Therefore, it is necessary to consider adjusting the dosage and course of treatment of this product.
