Maprotiline Hydrochloride Tablets

This product is a tetracyclic antidepressant. Its main function is to selectively inhibit the reuptake of norepinephrine in the peripheral and central nervous systems, while having no effect on the reuptake of 5-hydroxytryptamine. Due to the reduction of norepinephrine reuptake, the concentration of norepinephrine in the synaptic cleft increases, which downregulates the α2 receptors in the presynaptic membrane and strengthens the action of the α1 receptors in the postsynaptic membrane, producing an antidepressant effect. This product also has an anti-anxiety effect, and its sedative, anticholinergic and blood pressure-lowering effects are relatively mild.

The main ingredient of this product is maprotiline hydrochloride, and its chemical name is: N-methyl-9,10-ethanoanthracene-9(10H)-propylamine hydrochloride.

This product is white or off-white tablets.

It is used for various types of depression. It is also effective for post-schizophrenia depression.

Common oral dosage for adults: 25 mg once, 2 to 3 times a day, increase by 25 to 50 mg every other day according to the condition. The effective therapeutic dose is generally 75 to 200 mg a day, and the maximum dose should not exceed 225 mg a day. Be careful about the occurrence of adverse reactions. The maintenance dose is 50 to 150 mg a day, taken orally in 1 to 2 times.

Common anticholinergic symptoms include dry mouth, constipation, dysuria, dizziness, blurred vision and tachycardia, which are mild and often occur in the early stage of medication. Adverse reactions of the central nervous system may cause drowsiness, insomnia or agitation, and early medication may increase the risk of suicide in patients. Other symptoms include rash, postural hypotension and abnormal electrocardiogram changes, mainly conduction block. Epileptic seizures and toxic liver damage are occasionally seen.

Use with caution in patients with severe liver and kidney dysfunction, prostate hypertrophy, the elderly or cardiovascular disease. ECG should be monitored during use. This product should not be used in combination with monoamine oxidase inhibitors. This product should be used 14 days after monoamine oxidase inhibitors are discontinued. In the early stage of using this product, patients with suicidal tendencies should be closely monitored. If the patient has a tendency to turn manic, the drug should be stopped immediately. It is not advisable to drive a vehicle, operate machinery or work at height during medication.

Precautions for children: Forbidden for children under 6 years old, and children over 6 years old should refer to the adult dose and reduce the dose as appropriate. Precautions for pregnancy and lactation: Forbidden. Precautions for the elderly: Start with a small dose and slowly increase to an appropriate dose.

This product can weaken the antihypertensive effect of clonidine and guanethidine when used in combination with this product. 4. This product can increase arrhythmia when used in combination with thyroid hormones. 5. This product can increase the blood concentration of fluoxetine when used in combination with this product. It is not suitable to use it together.
