Cisapride dry suspension

The main ingredient of this product is cisapride, whose chemical name is (±)-cis-4-amino-5-chloro-N-[1-[3-(p-fluorophenoxy)propyl]-3-methoxy-4-piperidinyl]-2-methoxyformamide-hydrate

This product is white granules, odorless and sweet.

Severe gastrointestinal motility disorders that are intolerant to or ineffective with other treatments, such as chronic idiopathic or diabetic gastroparesis, chronic intestinal pseudo-obstruction, and gastroesophageal reflux disease

Oral treatment: Add water to 100ml before taking, shake well and take. This product should be taken 15 minutes before meals or before bedtime (if the 4th dose is needed). This product should be stored at 4-8℃ after adding water and used up within 7 days. This product should not be taken with grapefruit juice. Dosage: Adults: Depending on the severity of the disease, the total daily amount is 15mg-30mg, administered in 2-3 times. 5mg each time (the dose can be doubled). The maximum daily dose is 30mg. Children weighing 25kg-50kg: The maximum dose is 5mg, 4 times a day. Oral tablets or suspensions can be taken orally. The daily dose should not exceed 0.8mg/kg. Children weighing less than 25kg: 0.2mg/kg body weight each time, 3-4 times a day. It is best to use a suspension. The daily dose should not exceed

There are extremely rare (<1/10000) case reports of QT prolongation and/or serious (individually fatal) ventricular arrhythmias, such as torsade de pointes, ventricular tachycardia, and ventricular fibrillation. Most of these patients were treated with concomitant medications (including CYP3A4 enzyme inhibitors) and/or had heart disease or risk factors for arrhythmias. Other adverse reactions reported include: general adverse reactions (>1/100<1/10) (consistent with the pharmacological effects of cisapride), transient abdominal cramps, intestinal sounds, and diarrhea may occur. Less common adverse reactions (>1/1000<1/100) Occasionally allergic reactions (including rash, urticaria) occur.

Patients with known allergies to this product are contraindicated. Concomitant oral or parenteral use of drugs that strongly inhibit the CYP3A4 enzyme is prohibited, including: - triazole antifungal drugs; - macrolide antibiotics; - HIV protease inhibitors; - nefazodone; (see [Drug Interactions]) Patients with heart disease, arrhythmia, QT interval prolongation are contraindicated, and it is prohibited to use with drugs that cause QT interval prolongation; patients with water and electrolyte disorders, especially those with hypokalemia or hypomagnesemia; patients with bradycardia; patients with other serious cardiac rhythm diseases; patients with uncompensated heart failure; patients with congenital QT interval prolongation or a family history of congenital QT interval prolongation syndrome; patients with lung,

The main metabolic pathway of this product is through CYP3A4 enzyme metabolism. If drugs that inhibit this enzyme are used orally or parenterally at the same time, it may lead to increased plasma cisapride concentrations, thereby increasing the risk of QT interval prolongation and serious arrhythmias, including ventricular tachycardia, ventricular fibrillation and torsade de pointes.
