Eperisone Hydrochloride Tablets

Function and Efficacy

1. Relieve skeletal muscle hypertonia (1) Inhibit experimental spasticity. It can dose-dependently inhibit rat interthalamic transection spasticity (γ-tonicity) and ischemic cerebral spasticity (α-tonicity). (2) Inhibit spinal reflexes. In a spinal cat experiment, it can inhibit monosynaptic and polysynaptic reflex potentials caused by stimulation of the posterior roots of the cat spinal cord to the same extent. (3) Reduce the sensitivity of muscle spindles through the γ-system. It can inhibit the activity of afferent nerves (1α fibers) generated in human muscle spindles 30 minutes after administration. This product inhibits the spontaneous firing of animal γ-motor neurons, but does not directly act on muscle spindles. Therefore, it can be considered that this product reduces the sensitivity of muscle spindles with the help of the γ-system. 2. Dilate blood vessels and improve blood flow (1) Vasodilation can dilate blood vessels through calcium ion antagonism (rat pecking) and muscle sympathetic nerve inhibition (human). (2) Increase blood flow. In humans, monkeys and dogs, it can increase blood flow in the skin, muscles, external carotid artery, internal carotid artery and vertebral artery. 3. It has analgesic and pain reflex inhibitory effects. The results of the drug perfusing the spinal cord of rats to inhibit the pain reflex of tail pinch pain (TailPinch) and removing the drug showed that the drug showed analgesic effects at the spinal cord level. 4. It makes voluntary movements free. It can be used in cases of spastic paralysis such as stroke patients to improve the torque curve and electromyography of Cybex. Under the condition of reducing the strength of spastic muscles, the extension and bending movements of the upper and lower limbs are brisk and smooth, and voluntary movements are free. Toxicology study: Data show that long-term toxicity: SD rats were orally administered this product for 26 weeks or 52 weeks continuously, with a dose of 25.50, 100, and 200 mg/kg/day. No abnormalities were found in the 25 mg/kg group. Even the group above 50mg/kg did not have serious effects; beagle dogs were orally administered 10 and 25.625mg/kg/day for 53 consecutive weeks. No abnormalities were found in the 10mg/kg group. Although the group above 25mg/kg had vomiting, salivation, increased liver weight and mild hepatocyte enlargement, they recovered once the drug was discontinued. Reproductive toxicity test: SD rats were orally administered 25-500mg/kg/day before pregnancy and in the early stage of organ formation, during the perinatal period, during the lactation period, and rabbits during the organ formation period. The results showed that the maternal animals were affected when administered above 200mg/kg. Newborns in the perinatal and lactation period were found to have mild developmental delays, but no teratogenicity. There was no effect on the development of motor function, learning, reproductive function and F2 fetuses of newborns.
