Nisoldipine sustained-release capsules

Function and Efficacy

Nisoldipine is a dihydropyridine calcium channel antagonist (calcium ion antagonist or slow channel blocker) that inhibits the influx of calcium ions through the cell membrane into vascular smooth muscle and myocardial cells. It can reversibly compete with other dihydropyridine drugs for binding to calcium ion channels. Since the contraction process of vascular smooth muscle depends on the entry of extracellular calcium into myocytes through specific ion channels, the inhibition of calcium channels will lead to the dilation of arterioles. In vitro tests have shown that the effect of nisoldipine on the contraction process is selective, and its effect on vascular smooth muscle is stronger than that on myocardium. Although nisoldipine, like other dihydropyridine calcium channel blockers, has a negative inotropic effect in vitro, studies in anesthetized animals have shown that the dose of nisoldipine that causes vasodilation is lower than the dose that affects myocardial contractility.

Appearance

This product is a capsule containing yellow spherical pellets.

Indication

Primary mild to moderate hypertension.

Usage and Dosage

Oral administration: Adults take 1 tablet each time, once a day.

Adverse Reactions

In clinical trials conducted abroad, most patients tolerated it well. 10.9% of 921 patients taking SULAR (a nisoldipine sustained-release tablet) discontinued the drug due to adverse reactions, and 2.9% of 280 patients taking placebo discontinued the drug due to adverse reactions. The frequency of discontinuation due to adverse reactions was dose-related, with an incidence of 5.4% in the 10 mg/day group and 10.9% in the 60 mg/day group.

Precautions

Patients with hypersensitivity to dihydropyridine calcium channel blockers.

Drug Interactions

1. It has a synergistic effect when used with beta-blockers or other antihypertensive drugs, and attention should be paid to postural hypotension. 2. When used with cimetidine, the blood concentration of this product can be increased and the effect can be enhanced. 3. Quinidine may slightly reduce the area under the drug-time curve (AUC) of this product, and the dosage of this product may need to be adjusted. 4. Rifampicin accelerates the metabolism of this product and weakens the antihypertensive effect by inducing the activity of the metabolic enzyme of this product, so the dosage of this product needs to be adjusted.

Storage

Keep in a dark place, airtight container and at room temperature. The validity period is tentatively set at two years.

Packaging Specification
