Nimustine Hydrochloride for Injection
Function and Efficacy
1. Anti-tumor effect: (1) It shows high anti-tumor activity against mouse leukemia L-1210. (2) It has a wide anti-tumor spectrum against mouse transplanted tumors. It shows excellent effects on lymphocytic leukemia L1210, granulocytic leukemia C-1498, plasmacytoma X-5563 (ascites type), Ehrlich's ascites carcinoma, and dural sarcoma MS-147. (3) This agent has a life-extending effect on mice with lymphocytic leukemia L1210 and methylcholanthrene-induced malignant glioma cells transplanted into the brain. 2. Mechanism of action: The main mechanism of action is to alkylate intracellular DNA and reduce the molecular weight of DNA, thereby inhibiting DNA synthesis. Toxicology: 1. Acute toxicity: (1) LD50 value (mg/kg) When administered intravenously, the LD50 values of male and female mice are 65 and 59 mg/kg, respectively, and the LD50 values of male and female rats are 47.8 and 45.6 mg/kg, respectively. (2) Effects on blood (2.5, 5, 10 mg/kg intravenous administration in dogs) The red blood cell count and reticulocyte count of the 2.5 mg/kg group decreased on the 7th to 11th day of administration, and basically returned to normal on the 14th to 16th day. There was no significant change in the platelet count. The white blood cell count decreased significantly until the 7th day, but then gradually increased and returned to normal on the 35th day. The eosinophil count also had a similar trend. The red blood cell count and platelet count of the 5 mg/kg and 10 mg/kg groups decreased significantly, and all died on the 10th to 17th day. 2. Subacute toxicity: (0.4, 2, 10 mg/kg/day intravenous administration for 4 weeks in rats) 0.4 mg/kg group: Female rats had mild leukopenia, but the organ weight, especially the actual weight, did not change in accordance with the body weight ratio. 2 mg/kg group: There was inhibition of weight gain, atrophy of lymphatic system tissue and bone marrow, and leukopenia. 10mg/kg group: Death due to systemic bleeding tendency occurred (dogs were intravenously administered at 0.08, 0.2, 0.5mg/kg/day for 5 weeks). 3. Teratogenicity test (rat 0.05, 0.1, 0.5mg/kg/day, gestational day 7-17, intravenous administration) In late pregnancy, the average birth weight of the 0.5mg/kg group was reduced, and the group with more than 0.1mg/kg had polydactyly and other appearance abnormalities, which was teratogenic.
