Etoposide is chemically known as 9-(4, 6-o-ethylidene-beta-D-glucopyranoside)-4 '-demethylepipodophyllotoxin. Also known as etoposide, podophylloside, podophyllotoxin, podophylloside, etc., is a grayish white crystalline powder, odorless. It is easy to change color when exposed to light and heat, and has moisture-inducing properties. Almost insoluble in water, slightly soluble in methanol, dimethyl sulfoxide, slightly soluble in ethanol. Etoposide is a new semi-synthetic derivative of epipodophyllotoxin, a mitotic inhibitor, which can stop cells in the metaphase of mitosis and is a cell cycle specific anticancer drug. This product acts on DNA topoisomerase II (Topo II) to form a "drug chemicalbook-enzyme-DNA" complex, which blocks the repair of DNA by Topo II and leads to the obstruction of DNA replication, thus inhibiting the proliferation of tumor cells. It mainly acts on S phase and G2 phase cells and blocks cells in G2 phase. Experimental studies have found that the compound can be reversed with the elimination of the drug. Topo II is free, the damaged DNA can be repaired, and its anti-tumor effect is reduced, so prolonging the time of administration can improve the anti-tumor activity. It is mainly used in the treatment of small cell lung cancer, malignant lymphoma, malignant germinoma, leukemia, neuroblastoma, rhabdomyosarcoma, ovarian cancer, non-small cell lung cancer, gastric cancer and esophageal cancer.
