Pregabalin More Info

Pregabalin is a non-gamma-aminobutyric acid (GABA) receptor agonist or antagonist, a novel calcium channel modulator that blocks voltage-dependent calcium channels and reduces neurotransmitter release. It is used clinically for neuralgia associated with diabetic peripheral neuropathy, postherpetic neuralgia and adjunctive treatment of partial seizures in adults, generalized anxiety disorder, central neuralgia (including neuralgia accompanying spinal cord injuries, strokes and multiple sclerosis) and fibromyalgia.

Mechanism of Action

Pregabalin does not directly act on GABA receptors or convert to GABA. Instead, it binds to the alpha-2 delta subunit of voltage-gated calcium channels, thereby reducing the release of excitatory neurotransmitters such as glutamate and noradrenaline. This action helps calm overactive nerves and reduce pain signals sent by damaged nerves.

Uses

1. Epilepsy: Pregabalin is used as an adjunctive therapy for partial seizures with or without secondary generalization.
2. Neuropathic Pain: It is effective in treating various types of neuropathic pain, including diabetic peripheral neuropathy, postherpetic neuralgia, and other chronic pain conditions .
3. Fibromyalgia: Pregabalin is the first drug approved in the USA for the treatment of fibromyalgia.
4. Anxiety Disorders: It is also used to treat generalized anxiety disorder.

Dosage and Administration

Pregabalin is available in various forms including oral capsules, solutions, and extended-release tablets. The typical dosage ranges from 150 mg to 600 mg per day, divided into two or three doses. The starting dose is generally 150 mg daily, which can be adjusted based on the patient's response and tolerance
