2. Physical properties

Appearance: Yellow or orange yellow powder.

Melting point: 195 ℃ (decomposition).

Solubility: Slightly soluble in methanol, difficult to dissolve in water or ethanol, and cannot be completely dissolved with ether, acetone, or ethyl acetate, but easily soluble in DMSO (dimethyl sulfoxide).

3. Chemical properties

Thiazole blue is a diazonium salt with cell permeability and positive charge, which can be absorbed by cells through the plasma membrane and then reduced by NAD (P) H oxidoreductase inside the cell.

4. Purpose

Drug toxicity testing: used to determine the toxicity of drugs (including other treatment methods such as radiation exposure) to cells cultured in vitro.

Cell proliferation and activity assay: Succinate dehydrogenase in live cell mitochondria can reduce exogenous MTT to insoluble blue purple crystalline formazan and deposit it in cells, while dead cells do not have this function. Dimethyl sulfoxide can dissolve formazan in cells. Its light absorption value is measured at a wavelength of 490nm using an enzyme-linked immunosorbent assay (ELISA) reader. Within a certain range of cell numbers, the amount of MTT crystals formed is proportional to the number of cells. Based on the measured absorbance value, the number of live cells can be determined. The larger the OD value, the stronger the cell viability.

Dehydrogenase detection: can be used to detect the activity of intracellular dehydrogenases, thereby indirectly reflecting the metabolic status of cells.

Other applications: In the activity determination of liver cell regeneration factor, thiazole blue colorimetric method is used to determine the activity and specificity of liver cell regeneration stimulating factor; It can also be used to detect and evaluate the skin contact sensitization of chemicals.

5. Precautions

Storage conditions: It should be stored in the dark at 4 ℃ to ensure its stability and activity.

Solution preparation: MTT is generally best prepared on the spot, filtered and stored in the dark at 4 degrees Celsius for two weeks to be effective, or prepared at 5mg/ml and stored at -20 ℃ to avoid repeated freeze-thaw cycles. It is best to divide in small doses and wrap them in light proof bags or black paper or tin foil to prevent decomposition. When MTT turns gray green, it must not be used again.

Safe use: MTT is carcinogenic, be careful when using and wear gloves.
