Molecular targeted therapy drugs
The small molecule compound erlotinib, also known as erlotinib, itseva, tarceva, OSI-774, is a receptor tyrosine kinase inhibitor (EGFR antagonist), which belongs to molecular targeted therapy drugs. It inhibits the phosphorylation reaction by competing with ATP for the intracellular catalytic site of receptor tyrosine kinase within cells, thereby blocking the downstream proliferative signal transduction and inhibiting the ligand-dependent HER-1/EGFR activity of tumor cells, achieving the effect of inhibiting tumor cell proliferation. It is mainly used in clinical practice for locally advanced or metastatic non-small cell lung cancer (NSCLC) that is ineffective with other treatments, as well as for locally advanced unresectable or metastatic pancreatic cancer in combination with gemcitabine for first-line treatment. On November 2004, this drug was first approved in the United States for the treatment of locally advanced or metastatic non-small cell lung cancer (NSCLC) after at least one chemotherapy failure. In 2005, the results of a phase III clinical trial by the NCIC Canadian Clinical Trials Group indicated that gemcitabine combined with erlotinib (EGFR blocker) could increase the median survival time of advanced pancreatic cancer from 5.9 months to 6.4 months and the 1-year survival rate from 17% to 24%. On November 2005, Genentech and OSI jointly announced that the US Food and Drug Administration (FDA) approved erlotinib (tarceva) combined with gemcitabine as the first-line treatment for advanced pancreatic cancer.

Mechanism of action
Erlotinib is a highly specific anti-tumor targeted therapeutic drug developed by the British company AstraZeneca. It is the first molecular targeted drug used for the treatment of non-small cell lung cancer. It works by selectively inhibiting the signal transduction pathway of epidermal growth factor receptor tyrosine kinase (EGFR-TK).
