Description

Perphenazine is a phenothiazine compound that blocks dopamine D2 receptors and antagonizes H1, choline-like M1, and α1 adrenergic receptors in the vomiting center. Perphenazine induces cell death and mitochondrial damage, caspase-3 activation, and a decrease in intracellular ATP levels. Antioxidants inhibit perphenazine-induced cell death, while pan-capase inhibitors do not. Perphenazine at a concentration of 0.0001-0.01 μM had no significant effect on melanocyte viability. Treatment of these cells with higher concentrations of perphenazine will result in a concentration-dependent decrease in cell viability. EC50 is 2.76μM. 1-3 μM of Perphenazine also reduces tyrosinase activity and melanin content.
