1. Description

Corosolic acid is a naturally occurring pentacyclic triterpenoic acid found in a wide variety of plants, including Lagerstroemia grandiflora, loquat, apple, and basil. It appears as a white to off-white solid powder. It is soluble in DMSO (20–48 mg/mL), DMF (14 mg/mL), ethanol, and methanol, but is practically insoluble in water. It should be stored at –20°C in a light-protected, sealed container; in powder form, it has a shelf life of 2–3 years. This compound exhibits various biological activities, including antitumor, anti-inflammatory, antidiabetic, antioxidant, antihypertensive, and hypolipidemic effects.

2. Application

(1) Analytical Standards and Research Tools: Used as analytical standards for content determination, pharmacological experiments, and bioactivity screening, as well as for research applications such as cell-based drug delivery and animal studies.

(2) Oncology Research: Used to study the mechanisms of antiproliferation and apoptosis induction in various cancer cell lines, including hepatocellular carcinoma, lung cancer, colorectal cancer, cervical cancer, and leukemia. Studies indicate that it exerts antitumor activity through mechanisms such as inhibiting FAK signaling, inducing ERK-dependent apoptosis, and promoting β-catenin degradation.

(3) Antidiabetic Research: Used in animal models of type 2 diabetes to investigate the mechanisms underlying its hypoglycemic effects. Studies indicate that it improves sensitivity and inhibits postprandial hyperglycemia.

(4) Anti-inflammatory research: Used in acute and chronic inflammation models (such as ear edema) to investigate the mechanisms underlying its suppression of inflammatory responses.

3. Main Efficacy

(1) Antitumor activity: It exhibits cytotoxic effects against cancer cells such as HepG2, A549, SNU-C4, HeLa, S3, and K562, with half-maximal effective concentrations (EC₅₀) of 4.8, 5, 0.4, 1, and 4.3 μg/mL, respectively. It inhibits cancer cell growth through mechanisms such as inducing apoptosis, promoting N-terminal phosphorylation of β-catenin, and facilitating proteasomal degradation. It also exhibits inhibitory effects on colorectal and prostate cancer cells.

(2) Hypoglycemic Effects: In a KKAy mouse model of type 2 diabetes, doses of 2 and 10 mg/kg reduced blood glucose levels. It improves glucose metabolism and suppresses postprandial hyperglycemia, and is being studied for the management of type 2 diabetes.

(3) Anti-inflammatory activity: It inhibits furazolidone-induced ear edema in mice, with a half-maximal inhibitory concentration (IC₅₀) of 0.09 mg/ear.

(4) Cardiovascular protection: In a rat model of metabolic syndrome, a 0.072% dietary supplement reduced systolic blood pressure, lowered serum free fatty acid levels, and decreased oxidative stress markers (thiobarbituric acid reactive substances and 8-hydroxy-2'-deoxyguanosine), as well as myeloperoxidase markers (3-nitrotyrosine and 3-chlorotyrosine).

(5) Antioxidant Activity: It exhibits free radical scavenging and lipid peroxidation inhibitory activities, thereby alleviating oxidative stress-induced damage.

(6) Antihypertensive Activity: It demonstrates blood pressure-lowering effects in animal models of hypertension.

(7) Lipid-Lowering Activity: It reduces serum free fatty acid levels and improves lipid metabolism.

(8) Antimicrobial and Antiviral Activity: It possesses antifungal and antiviral activities.
