6-Methoxynicotinamide: Nicotinamide Analog with Anti-Inflammatory, Antidiabetic and Anticancer Activities as Multitarget Metabolic Modulator

1. Description

6-Methoxynicotinamide is an analog of nicotinamide and belongs to the pyridinecarboxamide class of compounds. It appears as a white to off-white solid powder. It is soluble in DMSO (≥60 mg/mL), has a molecular weight of approximately 152.15, and the molecular formula C₇H₈N₂O₂. Storage conditions are as follows: the powder can be stored for 3 years at -20°C and for 2 years at 4°C; when dissolved in a solvent, it can be stored for 6 months at -80°C. This compound is for research use only and must not be used for diagnosis or treatment.

2. Application

(1) NNMT Inhibition Studies: As a selective inhibitor of nicotinamide N-methyltransferase, it is used to investigate the mechanisms by which this enzyme regulates metabolism, provides cellular protection, and influences disease progression. The half-maximal inhibitory concentration (IC₅₀) for human NNMT is 1.8 μM.

(2) Metabolic Disease Research: Used in animal model studies of metabolic disorders such as obesity, type 2 diabetes, and non-alcoholic fatty liver disease to regulate glucose metabolism and body weight by inhibiting NNMT activity.

3. Main Efficacy

In a heart failure model, it significantly reduces the levels of 1-methylnicotinamide (by 77.1%) and its downstream metabolites in myocardial tissue.

(2) Regulation of glucose and lipid metabolism: In animal models of metabolic diseases, inhibiting NNMT activity improves glucose metabolism, regulates body weight, and exerts anti-obesity and anti-diabetic effects.

(3) Antitumor activity: In tumor cells such as osteosarcoma and Merkel cell carcinoma, it significantly reduces cell viability, increases reactive oxygen species (ROS) production, and activates apoptosis pathways.

(4) Cardiovascular protective effects: In a heart failure model with preserved ejection fraction, it improves cardiac function, alleviates left ventricular hypertrophy and fibrosis, reduces the expression of pro-inflammatory and pro-fibrotic genes, and decreases macrophage infiltration.
