    1. Description

    Apigenin is a naturally occurring flavonoid compound widely found in various plants, including parsley, chamomile, celery, spinach, and thyme. It appears as a yellow crystalline powder, is soluble in dimethyl sulfoxide and hot ethanol, and is practically insoluble in water. It should be stored in a cool, dry place, protected from light and kept in a sealed container. This compound exhibits various biological activities, including antioxidant, anti-inflammatory, antitumor, neuroprotective, and cardiovascular protective effects.

    2. Application

    (1) Oncology Research: Used to study the mechanisms of antiproliferation and apoptosis induction in various cancer cell lines, including breast cancer, colorectal cancer, liver cancer, lung cancer, prostate cancer, and osteosarcoma. It exerts antitumor activity by regulating multiple signaling pathways, such as PI3K/AKT, MAPK/ERK, JAK/STAT, NF-κB, and Wnt/β-catenin. Its ability to inhibit tumor growth has been demonstrated in in vivo animal models.

    (2) Neuroprotection Research: Used in studies of neurological disorders such as Alzheimer’s disease, Parkinson’s disease, and cerebral ischemia-reperfusion injury. Research indicates that it can improve cognitive dysfunction and alleviate histopathological damage in the hippocampus and thoracic aorta by inhibiting the AGEs/RAGE/NF-κB signaling pathway. In cerebral ischemia models, it exhibits antioxidant effects, reduces neuroinflammation, regulates apoptosis, and promotes neural regeneration.

    (3) Cardiovascular Disease Research: Used in studies of cardiovascular disease models such as myocardial infarction, cardiac hypertrophy, and myocardial ischemia-reperfusion injury.

    (4) Anti-inflammatory and Immunomodulatory Research: Used to investigate the mechanisms underlying the suppression of inflammatory responses. In lipopolysaccharide (LPS)-activated macrophages, apigenin significantly inhibits nitric oxide production (inhibition rate up to 98%), reduces superoxide anion production (inhibition rate 48.2%), and decreases hypochlorous acid production; its mechanism of action is associated with targeting the TLR4/MD-2 interface.

    (5) Osteoporosis Research: To investigate the mechanisms of bone-protective effects, including the promotion of osteoblast differentiation and the inhibition of osteoclast formation.

    (6) Metabolic Disease Research: For studies on hyperlipidemia, diabetes, and their complications. In a rat model of hyperlipidemia, apigenin reduces total cholesterol, triglyceride, and malondialdehyde levels while increasing superoxide dismutase activity; its antioxidant effects are mediated through the LDLR-LCAT signaling pathway.

    (7) Renal protection research: Used to study cisplatin- and doxorubicin-induced nephrotoxicity models as well as diabetic nephropathy models.

    3. Main Efficacy

    (1) Antioxidant effects: By scavenging free radicals, enhancing the activity of endogenous antioxidant enzymes, reducing malondialdehyde levels, and increasing the activity of superoxide dismutase and glutathione, it protects cells from oxidative stress damage. In FRAP, DPPH, and ABTS antioxidant assays, its free radical scavenging capacity exhibits a concentration-dependent response.

    (2) Anti-inflammatory effects: It inhibits the nuclear factor κB (NF-κB) signaling pathway and reduces the production of pro-inflammatory cytokines such as tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and interleukin-6 (IL-6). By targeting the TLR4/MD-2 interface to inhibit nitric oxide (NO) production, it alleviates inflammatory responses.

    (3) Antitumor activity: Inhibits cancer cell proliferation through various programmed cell death mechanisms, including apoptosis (intrinsic and extrinsic pathways), autophagy, necrotic apoptosis, and ferroptosis. It regulates the expression of apoptosis-related proteins such as p53, Bax/Bcl-2, and the caspase family.

    (4) Neuroprotective effects: Improves cognitive dysfunction by inhibiting the AGEs/RAGE/NF-κB signaling pathway. In cerebral ischemia models, it reduces neuroinflammation, inhibits apoptosis, and promotes neurogenesis.

    (5) Cardiovascular protective effects: By activating the deacetylase 3/β-catenin/PPARγ signaling pathway, it improves cardiac function after myocardial infarction and reduces myocardial cell apoptosis. It protects against isoproterenol-induced myocardial hypertrophy and myocardial infarction, alleviates doxorubicin-induced cardiotoxicity, and improves ischemia-reperfusion injury.

    (6) Anti-aging effects: By inhibiting the formation of advanced glycation end products (IC50 = 0.18 mM), reducing their binding to receptors, and downregulating the expression of phosphorylated nuclear factor κB, it delays D-galactose-induced aging and improves spatial learning and memory abilities.

    (7) Regulation of Glucolipid Metabolism: By activating the LDLR-LCAT signaling pathway, it reduces total cholesterol and triglyceride levels and mitigates oxidative damage induced by a high-fat diet. It exhibits protective effects in a hyperglycemia-dependent endothelial dysfunction model.

    (8) Bone-protective effects: By regulating levels of inflammatory factors and oxidative stress, it promotes osteoblast differentiation and inhibits osteoclast formation.

    (9) Organ-protective effects: It exerts protective effects on multiple organs, including the heart, brain, kidneys, and liver. It can alleviate cisplatin- and doxorubicin-induced nephrotoxicity and improve diabetic nephropathy and tubulointerstitial fibrosis.
