1. Description

Ecdysone is a naturally occurring steroid hormone belonging to the ecdysone family. It serves as a key regulatory hormone in the molting and metamorphosis processes of insects and other arthropods. It appears as a white to off-white solid powder, soluble in acetic acid, ethanol, methanol, or dimethyl sulfoxide; slightly soluble in chloroform; and insoluble in water. Its molecular formula is C₂₇H₄₄O₆, and its molecular weight is approximately 464.6. It should be stored at -20°C in a light-protected, sealed container. This compound is a precursor to 20-hydroxyecdysone, the primary ecdysone in insects, and exerts its effects through heterodimeric receptor complexes to regulate the expression of genes associated with molting and metamorphosis.

2. Application

(1) Research on insect development and metamorphosis: As a ecdysone receptor agonist, it is used to study the molecular mechanisms underlying developmental processes such as molting, metamorphosis, reproduction, and sleep in insects. Research indicates that ecdysone controls the synthesis of proteins required by larvae by regulating gene expression, and leads to the formation of polyline chromosomes (high-expression sites).

(2) Research on Gene Expression Regulation Systems: Used to develop controllable gene expression systems, enabling the spatiotemporal-specific expression of exogenous genes in scientific research, agriculture, and medicine through ecdysone-induced systems.

(3) Research on Cell Apoptosis and Renal Function: To investigate the mechanisms of cell apoptosis. Studies indicate that ecdysone (100 nM treatment for 48 hours) can induce apoptosis in medullary collecting duct cells within the renal tubules and, in a dose-dependent manner, increase the expression of the mesenchymal marker α-smooth muscle actin and the levels of cleaved caspase-3.

(4) Development of Environmentally Friendly Pesticides: Used to develop selective insect growth regulators as active ingredients in environmentally friendly pesticides. Ecdysone is found in various plants, primarily functioning as a defense mechanism (toxin or repellent) against herbivorous insects.

(5) Reproductive physiology research: Used to investigate the regulatory mechanisms of the insect reproductive cycle. Studies have shown that a pre-partum peak in ecdysone levels can induce mammary gland autophagy, promoting mammary gland regression, which is critical for maximizing the reproductive output of fruit flies.

3. Main Efficacy

(1) Ecdysone receptor agonist activity: As an endogenous ligand for the ecdysone receptor, it forms a heterodimeric receptor complex with hypervalvulin and binds to target gene DNA sequences to regulate the expression of genes associated with molting and metamorphosis.

(2) Induction of cell apoptosis: In medullary collecting duct cells of the renal tubules, ecdysone (100 nM) induces cell apoptosis, increases the expression of cleaved caspase-3, and induces a transition of cells from an epithelial phenotype to a mesenchymal-like morphology.

(3) Regulation of cell migration and proliferation: Studies have shown that ecdysone (10–100 nM) inhibits cell movement and scratch healing, and suppresses cell proliferation in a dose-dependent manner.

(4) Prohormone Function: As a precursor to 20-hydroxyecdysone, ecdysone is synthesized in the prothymus of insects, secreted into the hemolymph, and enzymatically converted into the more active 20-hydroxyecdysone in target tissues.

(5) Cross-species biological activity: In addition to insects, ecdysone exhibits biological activity in mammalian cells, where it activates mineralocorticoid receptors and induces apoptosis. In certain plants, ecdysone acts as a chemical defense compound, exerting anti-feeding and toxic effects on herbivorous insects.
