D-Glucose 6-phosphate

D-Glucose 6-phosphate (G6P), often called the Robison ester, is a fundamental phosphorylated sugar that functions as the ultimate gateway and command center for cellular carbohydrate metabolism. Positioned at the very start of intracellular glucose utilization, it dictates whether glucose is broken down for immediate energy, stored for later, or used to build cellular defenses.

• Gateway to Glycolysis: After glucose enters a cell, it is immediately phosphorylated into G6P by hexokinase or glucokinase. This traps the glucose inside the cell. G6P is then isomerized into D-fructose-6-phosphate (F6P) by phosphoglucose isomerase to fuel the glycolytic pathway for ATP production.
• Initiator of the Pentose Phosphate Pathway (PPP): G6P acts as the direct substrate for glucose-6-phosphate dehydrogenase (G6PD), the rate-limiting step of the PPP. This pathway is the cell’s primary source of NADPH (crucial for fighting oxidative stress and synthesizing fatty acids) and ribose-5-phosphate (the building block for DNA and RNA).
• Precursor for Glycogen Synthesis: When blood sugar and cellular energy are high, G6P is converted by phosphoglucomutase into glucose-1-phosphate. This intermediate is then used to build glycogen, archiving the glucose for future fasting states.
• Terminal Step of Gluconeogenesis and Glycogenolysis: In the liver and kidneys, the enzyme glucose-6-phosphatase removes the phosphate group from G6P. This converts it back into free glucose, allowing it to be released into the bloodstream to maintain systemic blood sugar levels.
