SS-31 (Elamipretide, also known as MTP‑131 or D‑Arg‑Dmt‑Lys‑Phe‑NH₂) is a synthetic tetrapeptide composed of four amino acids: D‑arginine, 2’,6’‑dimethyltyrosine (Dmt), L‑lysine, and L‑phenylalanine. The presence of D‑amino acids at the first two positions confers enhanced stability against enzymatic degradation. The molecular formula of the free base is C₃₂H₄₉N₉O₅, and its molecular weight is approximately 639.79 g/mol (free base) or 698.78 g/mol (acetate salt).

SS-31 is designed to target the inner mitochondrial membrane by binding to cardiolipin, a phospholipid essential for the structural integrity of the electron transport chain.
