    Product Overview

    Palbociclib is a potent, orally bioavailable cyclin-dependent kinase 4/6 (CDK4/6) inhibitor used in the treatment of hormone receptor-positive (HR+), human epidermal growth factor receptor 2-negative (HER2-) advanced or metastatic breast cancer. Marketed under the brand name Ibrance, palbociclib was the first CDK4/6 inhibitor approved by the FDA in February 2015, representing a major breakthrough in breast cancer therapy. By selectively inhibiting CDK4/6 kinases, palbociclib blocks cell cycle progression from G1 to S phase, thereby inhibiting tumor cell proliferation.

    Chemically, palbociclib is 6-acetyl-8-cyclopentyl-5-methyl-2-[[5-(piperazin-1-yl)pyridin-2-yl]amino]pyrido[2,3-d]pyrimidin-7(8H)-one with the molecular formula C₂₄H₂₉N₇O₂ and a molecular weight of 447.53 g/mol. It is a yellow to orange-yellow crystalline powder with solubility in DMSO and DMF. The compound exhibits high selectivity for CDK4 and CDK6 over other CDKs and favorable pharmacokinetic properties including excellent oral bioavailability.
