Advantame (CAS 714229-20-6) is a high-intensity, non-caloric artificial sweetener derived from aspartame and vanillin . Structurally, it is an N-[N-[3-(3-hydroxy-4-methoxyphenyl)propyl]-L-α-aspartyl]-L-phenylalanine 1-methyl ester monohydrate, combining the peptide backbone of aspartame with a vanillyl substituent .

With a purity of ≥98.0%, Advantame is reported to be approximately 20,000 times sweeter than sucrose by weight and about 110 times sweeter than aspartame . This exceptional sweetness potency makes it effective at very low concentrations, contributing minimal calories to formulations.

Synthetic Process

The synthesis of Advantame involves a multi-step process :

1. Step 1: Production of the principal manufacturing intermediate, 3-hydroxy-4-methoxycinnamaldehyde (HMCA)

2. Step 2: Hydrogenation to form 3-(3-hydroxy-4-methoxyphenyl) propionaldehyde (HMPA)

3. Step 3: The HMPA methanol solution is combined with aspartame to form the imine, followed by selective hydrogenation to yield Advantame

Careful protection and deprotection strategies are employed to ensure selective reaction at the desired functional groups, minimizing racemization and side reactions . The final product is purified by crystallization to achieve high purity suitable for food applications

Mechanism of Action

Advantame exerts its sweet taste by binding to sweet taste receptors (T1R2/T1R3) on the tongue. Its unique structure—combining the aspartame dipeptide backbone with a vanillyl group—enhances sweetness potency and resistance to hydrolysis . Unlike aspartame, which can degrade under heat, Advantame maintains sweetness during cooking or storage, making it suitable for baked goods and acidic beverages .

Safety & Metabolism: Advantame is largely metabolized to amino acids and vanillin derivatives in the digestive tract, with minimal systemic exposure to the intact molecule .
