Product Overview

Indirubin (CAS 479-41-4) is a bisindole alkaloid and the main bioactive component of the traditional Chinese medicine Indigo naturalis . It is a structural isomer of indigo, distinguished by an asymmetric double bond between the C2 position of one indole ring and the C3 position of the second, giving it a characteristic red-purple color .

With a purity of ≥98.0%, Indirubin is a potent inhibitor of cyclin-dependent kinases (CDKs) and glycogen synthase kinase-3β (GSK-3β). It has been used in traditional Chinese medicine (Danggui Longhui Wan) for the treatment of chronic myelocytic leukemia (CML) since the 1970s, representing a significant discovery in Chinese medicine research

Synthetic Process

Indirubin can be obtained through natural extraction from indigo-containing plants or via chemical synthesis. The most representative chemical synthesis involves an aldol condensation reaction between isatin and indoxyl or acetyl indoxyl . An alternative one-step synthesis uses reductive coupling of isatin with KBH₄ to produce indirubin derivatives . Additionally, biocatalytic methods using enzymes such as toluene-4-monooxygenase (T4MO) have been developed for eco-friendly biosynthesis

Mechanism of Action

Indirubin exerts its anticancer effects primarily through the inhibition of cyclin-dependent kinases (CDKs) and GSK-3β, regulating the expression of Bax, Bcl-2, C-MYC, and matrix metalloproteinases (MMPs) via the PI3K/AKT/mTOR, NF-κB, MAPK, and JAK/STAT3 signaling pathways . It inhibits DNA synthesis in various cell lines and has been shown to reduce leukemia cell proliferation .

Beyond anticancer activity, indirubin exhibits anti-inflammatory properties by suppressing LPS-induced NF-κB and MAPK signaling pathways, reducing the expression of TNF-α, IL-6, and other pro-inflammatory cytokines. It also shows neuroprotective and antibacterial potential
