  • Description
Retatrutide functions as a first-in-class triple hormone receptor agonist (often nicknamed "Triple G").
Unlike previous generations of weight-loss drugs that target one or two metabolic pathways, retatrutide simultaneously activates three different nutrient-sensing hormone receptors: GLP-1, GIP, and Glucagon.

    The Triple-Action Mechanism
By mimicking three naturally occurring gut and pancreatic hormones, retatrutide targets appetite, glucose homeostasis, and energy expenditure simultaneously:

    1. GLP-1 (Glucagon-Like Peptide-1) Receptor Agonism
    • Suppresses Appetite: Acts on the brain's hypothalamus to significantly lower hunger levels and increase satiety (the feeling of fullness).
    • Slows Digestion: Delays gastric emptying, keeping food in the stomach longer to reduce immediate calorie intake.
    • 2. GIP (Glucose-Dependent Insulinotropic Polypeptide) Receptor Agonism
      • Optimizes Insulin Release: Enhances glucose-dependent insulin secretion from the pancreas, lowering blood sugar spikes after meals.
      • Improves Fat Metabolism: Works synergistically with GLP-1 to minimize nausea while improving the body's sensitivity to insulin and handling of lipids.

      3. Glucagon Receptor Agonism (The Differentiating Factor)
      • Increases Energy Expenditure: While traditional glucagon spikes blood sugar during fasting, pharmaceutical agonism here prompts the body to burn more calories at rest.
      • Accelerates Lipolysis: Actively signals the liver and adipose tissues to break down stored fat into usable energy.
    ECHEMI Editorial Reference
Retatrutide is a novel triple agonist peptide of the glucagon receptor (GCGR), glucose-dependent insulinotropic polypeptide receptor (GIPR) and glucagon-like peptide-1 receptor (GLP-1R). Retatrutide inhibits human GCGR, GIPR and GLP-1R with EC50 values of 5.79, 0.0643 and 0.775 nM, respectively and mouse GCGR, GIPR, and GLP-1R with EC50 values of 2.32, 0.191 and 0.794 nM, respectively. It is an important tool for obesity research.Retatrutide potently activates the GLP-1R signaling pathway to stimulate glucose-dependent insulin secretion through activity at the GIP receptor (GIPR) or the GLP-1R.Retatrutide is a synthetic peptide with glucose-lowering effects. It is an antidiabetic agent against type 2 diabetes (T2D), stimulating insulin and suppressing glucagon secretion in a glucose-dependent manner.Retatrutide was also shown to delay gastric emptying, lower fasting and postprandial glucose concentration, decrease food intake and reduce body weight in patients with type 2 diabetes.
