Description:
5-Amino-1MQ is a selective inhibitor of nicotinamide N-methyltransferase (NNMT) with an IC₅₀ of 1.2 μM. It exhibits 10-fold higher potency compared to unsubstituted 1-MQ (IC₅₀ = 12.1 μM) and 7.5-fold higher than the endogenous product 1-MNA (IC₅₀ = 9.0 μM). This compound demonstrates excellent membrane permeability (PAMPA flux = 3.01×10⁻⁶ cm/s), ensuring intracellular target engagement. By inhibiting NNMT activity, 5-Amino-1MQ reduces the conversion of nicotinamide to 1-methylnicotinamide, thereby elevating intracellular NAD⁺ levels, activating the SIRT1 pathway, and promoting energy metabolism and lipolysis. In animal studies, no adverse effects were observed at doses up to 60 mg/kg/d, demonstrating a favorable safety profile. It is widely used in research on metabolic regulation, lipogenesis inhibition, NAD⁺ enhancement, muscle regeneration, and anti-aging.
