Dabogratinib (TYRA-300) is an orally bioavailable, highly selective fibroblast growth factor receptor 3 (FGFR3) tyrosine kinase inhibitor designed to target oncogenic FGFR3 alterations, including activating mutations, FGFR3-TACC3 fusions, and gatekeeper resistance mutations such as V555L/M. By binding to the FGFR3 kinase domain and inhibiting its signaling activity, dabogratinib suppresses downstream pathways including ERK/MAPK signaling, thereby reducing FGFR3-driven tumor cell proliferation and survival. Its high selectivity for FGFR3 over FGFR1, FGFR2, and FGFR4 may help minimize effects associated with broader FGFR inhibition. Preclinical studies demonstrate potent activity against FGFR3-driven bladder cancer models and maintained activity against the FGFR3 V555M resistance mutation. Potential applications of dabogratinib include research on FGFR3-mutant and FGFR3-fusion-positive cancers, particularly metastatic urothelial carcinoma and bladder cancer, as well as studies of FGFR3-mediated oncogenic signaling, drug resistance, and targeted cancer therapy.
