    Dermorphin Usage And Synthesis
Structure Dermorphin does not contain the common N-terminal sequence for the traditional endogenous  peptides (Tyr–Gly–Gly–Phe), and its sequence is completely different from those of the endomorphins, which were identified as endogenous μ- peptides that also do not contain the common sequence. Interestingly, DM contains D-alanine (a D-isomer amino acid) in its sequence. The primary structure of dermorphin is YDAFGYPS-NH2.
Description Dermorphin was first isolated from the skin of the skin of South American frogs of the genus Phyllomedusa.  It is a high potency natural opiate peptide that binds selectively to mu- receptors.
    Uses A mu- receptor agonist
Definition ChEBI: Dermorphin is an oligopeptide.
Side effects Like μ-opiate agonists, dermorphins produce antinociception and also catalepsy, respiratory depression, constipation, tolerance, and dependence, although at a lower degree than   does[1].
in vitro Dermorphin, a peptide isolated from the skin of Phyllomedusa frogs and the peptide receptor (NOP) component by the endogenous agonist nociceptin/orphanin FQ (N/OFQ). In displacement binding studies at CHOhMu, Dermorphin and DeNo displac the binding of [3H]-DPN in a concentration dependent and saturable manner. Dermorphin displays an affinity of 7.17, while N/OFQ fails to displace [3H]-DPN at the Delta receptor. Dermorphin and DeNo stimulate the binding of GTPγ[35S] in a concentration dependent and saturable manner at the Mu receptor.

    ECHEMI Editorial Reference
Erdafitinib (JNJ-42756493) is a potent and orally available FGFR family inhibitor; inhibits FGFR1/2/3/4 with IC50s of 1.2, 2.5, 3.0 and 5.7 nM, respectively.
In early April of 2019, the US FDA approved Janssen Pharmaceutical Companies' brand name Balversa (erdafitinib) as the first-ever fibroblast growth factor receptor (FGFR) kinase inhibitor indicated for patients with locally advanced or metastatic urothelial carcinoma, with susceptible FGFR3 or FGFR2 genetic alterations, that has progressed during or following platinum-containing chemotherapy, including within 12 months of neoadjuvant or adjuvant platinum-containing chemotherapy. At the same time, the FDA also approved the therascreen FGFR RGQ RT-PCR Kit (Qiagen) for utilization as a companion diagnostic with erdafitinib for selecting patients for the indicated therapy. Erdafitinib's innovation lies in the fact that it is the first personalized treatment targeting susceptible FGFR genetic alterations for patients with metastatic bladder cancer, which demonstrates the design of erdafitinib in developing more personalized and precision medicines with the capacity to target cancer treatment to a patient's specific genetic mutation. Considering urothelial cancer is statistically the fourth most common kind of cancer in the world, the introduction of erdafitinib offers a welcome new option in the ever-expanding therapeutic tool kit to treat such prevalent medical conditions. Nevertheless, although erdafitinib was granted Breakthrough Therapy designation and Accelerated Approval from the FDA so as to allow the agency to focus on and expedite the approval process for a medication indicated for a serious condition that fills an unmet medical need using clinical trial data that is believed to predict a genuine clinical benefit for patients with the given condition, such designations mean further ongoing clinical trials are necessary to confirm the clinical benefit of erdafitinib going forward.|Erdafitinib is an oral small molecule inhibitor of fibroblast growth factor receptors (FGFR) 1 to 4 that is used in the therapy of locally advanced, unresectable or metastatic urothelial carcinoma. Erdafitinib has been associated with a high rate of serum enzyme elevations during therapy, but has not been linked to cases of clinically apparent acute liver injury.|Erdafitinib is an orally bioavailable, pan fibroblast growth factor receptor (FGFR) inhibitor with potential antineoplastic activity. Upon oral administration, erdafitinib binds to and inhibits FGFR, which may result in the inhibition of FGFR-related signal transduction pathways and thus the inhibition of tumor cell proliferation and tumor cell death in FGFR-overexpressing tumor cells. FGFR, upregulated in many tumor cell types, is a receptor tyrosine kinase essential to tumor cell proliferation, differentiation and survival.

Pan-FGFR tyrosine kinase inhibitor (FGFR1-4); antineoplastic (ATC L01EN01)

    Status:

Approved (FDA accelerated approval 2019-04-12; converted to regular approval 2024-01-19; EMA approval 2024-08)

    Mechanism:

Erdafitinib is an orally bioavailable, functionally selective small-molecule inhibitor of fibroblast growth factor receptors 1-4 (FGFR1-4). It inhibits FGFR tyrosine kinase activity (IC50 1.2-5.7 nM), blocking downstream RAS-MAPK/ERK and PI3K-AKT signaling that drives proliferation and survival of tumor cells harboring activating FGFR alterations (mutations, amplifications, or fusions). Rapid lysosomal uptake contributes to prolonged inhibition of FGFR signaling.
