2,6-Dibromo-3-nitropyridine Use and Manufacturing

Intermediate 6: 2, 6-dibromo-3-nitropyridine (Scheme 12); A mixture of commercially available 2, 6-dichloro-3-nitropyridine (10.0 g; 51.8 mmol) and 33 wpercent HBr/AcOH (120 mL) is heated at 80°C for 3h. The solution is concentrated in vacuo, the resulting residue is taken into EtOAc and ished with saturated aqueous sodium bicarbonate. The organic phase is dried over sodium sulfate and concentrated in vacuo. The resulting product 14.4 g (99percent) is used without further purification (Intermediate 6). GC/MS: 94percent purity, tStep 1. Preparation of 2, 6-dibromo-3-nitropyridine:2, 6-dichloro-3-nitropyridine (lOg, 51.8mmol) was taken up in a 40 percent hydrogen bromide in glacial acetic acid solution (lOOmL) and the resulting mixture was stirred at 80 °C for 18 h. Upon cooling to room temperature, the reaction mixture wasconcentrated under reduced pressure. The resulting residue was taken up in ethylacetate (EtOAc) and washed with saturated aqueous sodium bicarbonate. The organic phase was dried (Na2, 6-Dibromo-3-nitropyridine Heat at 50 °C a mixture of 2, 6-dichloro-3-nitropyridine (9 g) and hydrobromic acid (90 mL) in AcOH (30percent) overnight. Cool to RT and pour into water (600 mL). Filter the solid to provide the title compound (87percent).
