Toxicity
/AQUATIC SPECIES/ ... In the present study, the stereoisomeric separation and toxicity of fosthiazate, a chiral OP with two asymmetric centers on phosphorus and carbon atoms, was investigated. ... The inhibition potencies of the stereoisomers against acetylcholinesterase of Electrophorus electricus were slightly stereoselective, with a maximum difference of 1.4-fold among the isomers. A 3.1-fold difference, however, was observed in the acute toxicity of isomers to Daphnia magna. The 48-hr toxicity of isomers to D. magna followed the order pk1 > pk2 > pk4 > racemate approximately pk3. The stereoselective toxicity to D. magna found in fosthiazate suggests that the environmental safety of fosthiazate should be evaluated on the basis of its individual isomers.
Fosthiazate's production may result in its release to the environment through various waste streams; its use as a nematicide/insecticide(1) will result in its direct release to the environment(SRC).
TERRESTRIAL FATE: Based on a classification scheme(1), an estimated Koc value of 71(SRC), determined from a log Kow of 1.68(2) and a regression-derived equation(3), indicates that fosthiazate is expected to have high mobility in soil(SRC). Mobility is subject to pH and organic matter content(4). The compound was shown to be more persistent in soils with a pH of <6, exhibiting half-lives ranging from 53.3 to 57.7 days versus 14.1 to 20.7 days for soils with pH >7(5). Volatilization of fosthiazate from moist soil surfaces is not expected to be an important fate process(SRC) given a Henry's Law constant of 1.7X10-10 atm-cu m/mole(2). Fosthiazate is not expected to volatilize from dry soil surfaces(SRC) based upon a vapor pressure of 4.20X10-6 mm Hg at 25 °C(2).|TERRESTRIAL FATE: Under terrestrial field conditions, fosthiazate exhibited a dissipation half-life of 10-17 days. Vapor-phase fosthiazate is degraded in the atmosphere by reaction with photochemically-produced hydroxyl radicals(SRC); the half-life for this reaction in air is estimated to be 1.5 hours(SRC), calculated from its rate constant of 8.76X10-11 cu cm/molecule-sec at 25 °C(SRC) that was derived using a structure estimation method(3). Particulate-phase fosthiazate may be removed from the air by wet or dry deposition(SRC). Fosthiazate contains chromophores that absorb at wavelengths >290 nm(4), and therefore may be susceptible to direct photolysis by sunlight(SRC).
The rate constant for the vapor-phase reaction of fosthiazate with photochemically-produced hydroxyl radicals has been estimated as 8.76X10-11 cu cm/molecule-sec at 25 °C(SRC) using a structure estimation method(1). This corresponds to an atmospheric half-life of about 1.5 hours at an atmospheric concentration of 5X10+5 hydroxyl radicals per cu cm(1). Fosthiazate exhibits rapid hydrolysis in the environment under alkaline conditions with a half-life of <7 days reported. Under neutral conditions, hydrolysis is slow as suggested by a half-life of 104 days(2). Fosthiazate contains chromophores that absorb at wavelengths >290 nm(3), and therefore may be susceptible to direct photolysis by sunlight(SRC). An estimated BCF of 6 was calculated in fish for fosthiazate(SRC), using a log Kow of 1.68(1) and a regression-derived equation(2). Using a modeling approach based on the quantitative structure-activity relationship (QSAR), a log BCF of 0.50515, corresponding to a BCF of 3 was calculated(3). According to a classification scheme(3), these BCF values suggest the potential for bioconcentration in aquatic organisms is low(SRC).
The Koc of fosthiazate is estimated as 71(SRC), using a log Kow of 1.68(1) and a regression-derived equation(2). According to a classification scheme(3), this estimated Koc value suggests that fosthiazate is expected to have high mobility in soil. The compound is mobile in soil and can leach in acidic soils with low organic matter content as indicated in soil column studies. Using a soil column test employing three orchard soils from Veris, Greece collected in April 2006, fosthiazate was detected in leachates at 0.04, 0.09, and 0.004% of applied on clay (1.77% organic matter, pH 8.10, 37.0% sand, 21.0% silt, 41.0% clay), sandy clay loam (0.49% organic matter, pH 4.80, 57.0% sand, 21.0% silt, 22.0% clay), and clay loam (1.58% organic matter, pH 8.59, 43.0% sand, 19.0% silt, 38.0% clay, respectively. The pH increased with increasing soil depth while organic matter content decreased with increasing soil depth. Dissipation due to biotic and abiotic processes accounted for loss of 97.97%, 94.15%, and 99.62% of applied(4). Freundlich adsorption coefficients (Kd) in three contrasting US soils were determined. Kd values of 0.55 in San Emigdio sandy loam (12% clay, 12.5% sand, 75.4% silt, 0.45% organic matter, pH 7.2), 0.10 in Arlington sandy loam (9% clay, 67% sand, 24% silt, 0.82% organic matter, pH 6.7), and 0.89 in Waukegan clay loam (21% clay, 41% sand, 38% silt, 0.3.1% organic matter, pH 5.5) were measured(5). Freundlich adsorption coefficients of 1.23 to 2.74 mL/g indicate generally weak adsorption and adsorption is shown to be strongly correlated with soil organic matter content(6).
The Henry's Law constant for fosthiazate is 1.32X10-7 atm-cu m/mole(1). This Henry's Law constant indicates that fosthiazate is expected to be essentially nonvolatile from water and moist surfaces(2). Fosthiazate is not expected to volatilize from dry soil surfaces(SRC) based upon a vapor pressure of 4.2X10-6 mm Hg(1).
Fosthiazate was detected at a range of 4.0 to 12.2 ng/g in 4 of 173 samples of Japanese agricultural products (carrots, bananas, grapefruit); limit of detection 1.6 ng/g(1).
Occupational exposure to fosthiazate may occur through inhalation and dermal contact with this compound at workplaces where fosthiazate is produced or used. Limited data indicate that the general population may be exposed to fosthiazate via ingestion of contaminated food. (SRC)

Groups of 5 Crl:CD BR VAF/Plus rats/sex per group were dosed daily for 14 days by gavage (in 2 mL/kg corn oil vehicle) with fosthiazate at 2 mg/kg. Rats were then administered 2 mg/kg labeled fosthiazate, and were killed after 24 hrs or 168 hrs, at which time tissues (including blood) were collected for radiolabel analyses. Urine, feces, and expired air were collected at intervals after dosing. Unlabeled fosthiazate was 99% pure, and ring-labeled fosthiazate was >99% pure. There was no apparent effect of pre-treatment on metabolism... Patterns of excretion and tissue residues over time were comparable to single dose treatment. Fosthiazate is rapidly absorbed, metabolized, and excreted. Urine was the major route of excretion, whereas feces and expired air were minor routes. Neither sex-related differences in route or rate of excretion were observed. Excretion was essentially complete by 24 hrs after dosing. At 24 hrs, 75 to 82% of administered dose was eliminated. By 168 hrs after termination, 70% and 13.2% of administered dose was excreted in urine and feces, respectively. About 5.3% was eliminated as CO2.|Groups of 5 Crl:CD BR VAF/Plus rats/sex per group were dosed once by gavage (in 2 mL/kg corn oil vehicle) with fosthiazate, either 2 or 20 mg/kg. Unlabeled fosthiazate was 99% pure, and ring-labeled fosthiazate was >99% pure. Excreta and exhaled air were sampled throughout the 7-day post-exposure period. Major tissues were examined at termination for label. Expired air contained 5-6% of administered dose (in 1 N NaOH traps, presumed to be CO2), regardless of sex or dose level. Most of this was collected in the first 12 hrs. Amounts of volatile organics collected downstream from the CO2 traps were minuscule. Urinary metabolites accounted for 66-67% of administered dose in males, and 71- 73% in females. There was no apparent difference between the dose levels. Over one-half of the urinary label was collected within 6 hrs of dosing in all cases. Fecal metabolites constituted 11-12% of administered dose, regardless of sex or dose. Most of this label was collected within 48 hrs. Total percentage of administered dose found in all body tissues combined were 11.0 and 9.4% in 2 mg/kg/day and 20 mg/kg/day males, respectively; and 8.2 and 6.8% in respective females, suggesting appreciable retention. Tissue label was widely distributed at 1 week post-treatment. Highest concentrations of label at 2 mg/kg were in liver, lung, and heart. Radioactivity was clearly not concentrated in points of entry (g.i. tract) or circulation (blood), nor in the fat. This appreciable tissue retention, coupled with the significant CO2 production of labeled carbon from metabolism of the ring component, suggests that carbon from the thiazolidinyl ring is substantially assimilated into the body's carbon pool. ...|Ten Crl:CD BR VAF/Plus rats/sex/group were dosed once daily by gavage (in 2 mL/kg corn oil vehicle) with unlabeled fosthiazate at 2 mg/kg/day. Groups of 7 Crl:CD BR VAF/Plus rats/sex were dosed once by gavage (in 2.5 mL/kg corn oil vehicle) with fosthiazate at 18 mg/kg. Non-radiolabeled fosthiazate, which contained 13C on 50% of each of the methyl carbons of the butyl group, was 99.3% pure. S-sec-butyl-2-14C-labeled fosthiazate was 97.7% pure (by MS). Investigators evaluated fate of fosthiazate in tissues and excreta over 48 hrs, and characterized major metabolites found in urine. S-sec-butyl-2-14C-labeled group in the present study produced about the same distribution of label in excreta as had been observed in /another study/ which utilized ring-labeled 14C. This suggests that both the ring and the butyl substituents undergo degradation to release appreciable CO2 (about 10% of administered dose). Identified large MW metabolites (excluding glutathione products) underwent ring opening, often with subsequent methylation of the ring sulfur. Products of oxidation of this ring sulfur, such as sulfonic acids, sulfoxides, and sulfones, constituted about 20% of administered dose. There were several observed residues of hydrolysis of the S-sec-butyl group, similarly displaying oxidation of the sulfur, with or without methylation of the sulfur. ... The inhibition potencies of the stereoisomers against acetylcholinesterase of Electrophorus electricus were slightly stereoselective, with a maximum difference of 1.4-fold among the isomers. A 3.1-fold difference, however, was observed in the acute toxicity of isomers to Daphnia magna. The 48-hr toxicity of isomers to D. magna followed the order pk1 > pk2 > pk4 > racemate approximately pk3. The stereoselective toxicity to D. magna found in fosthiazate suggests that the environmental safety of fosthiazate should be evaluated on the basis of its individual isomers.
