Thrombin Use and Manufacturing

Methods of Manufacturing

Raw material processing and precipitation Take 2kg of pig blood, add 200ml of sodium citrate (38g/L or 3.8%) solution for anticoagulation, mix well, separate out, take out the supernatant plasma, add 10 times the amount of distilled water to the plasma Dilute, adjust Ph to 5.3 with 300ml of acetic acid (1%), after the precipitation is complete, centrifuge in a centrifuge for 5-10 minutes, discard the centrifugal fluid, collect the precipitate to obtain prothrombin. Animal blood sodium citrate → raw material treated plasma distilled water: HAc; Ph5.3 → precipitation prothrombin separation, precipitation, washing to take prothrombin, at 25-30C0, add 700ml of sodium chloride (0.9%) solution, Dissolve it, then add 15g calcium chloride to each liter of solution, stir well for 10min to dissolve it, leave it in the cold room for 1h, centrifuge, take the supernatant and add the same amount of frozen acetone, stir well and let it stand overnight. After centrifugation, the precipitate is finely ground with frozen acetone, placed in a cold room for 2 to 3 days, filtered to obtain the precipitate, washed with ether, and dried in vacuum to obtain crude thrombin, about 10 g. Prothrombin NaCl; CaCl2; acetone→separation; precipitation precipitation ether→washing crude thrombin to remove impurities, precipitation and drying. Add 200ml sodium chloride solution (9g/L) to the crude thrombin to dissolve it. Place it at 00C for 6 hours, filter, take the precipitate and add 150ml sodium chloride solution (9g/L) to repeat the above operation once, and combine the two filtrates. Adjust Ph to 5.5 with acetic acid (1%). Centrifugal separation, precipitation discarded, take the supernatant and add 2 times the amount of frozen acetone to the supernatant, let it stand for 2 hours, after the precipitation is complete, centrifuge, the precipitate is finely ground with frozen acetone, placed for 24 hours, and the solution is filtered. The precipitate was washed with absolute ethanol and ether, and dried in vacuum to obtain refined thrombin, about 0.4 g. Crude thrombin NaCl; HAc; ^0<0C; pH5.5 → filtrate acetone → precipitation and dried refined products
