Acid Blue 83 Use and Manufacturing

Methods of Manufacturing

Using p-chlorobenzaldehyde, 1-(N-ethylaniline) m-toluenesulfonic acid and p-phenetidine as raw materials, first p-chlorobenzaldehyde and 1-(N-ethylaniline) Condensate with m-toluenesulfonic acid, then oxidize the condensation product with potassium dichromate-oxalic acid, and finally condense with p-aminophenyl ether to obtain the product. After salting out, filtering, drying and crushing, the finished product is obtained. . After beating 1-(N-ethylaniline) m-toluenesulfonic acid with water, adjust the pH value to about 3, add p-chlorobenzaldehyde, and raise the temperature to 95-98°C to react to the end point. After distilling off excess p-chlorobenzaldehyde, dilute with water, lower the temperature, and add potassium dichromate, sulfuric acid and oxalic acid below 10°C for oxidation. At the end of the reaction, sulfuric acid was added at 45°C to precipitate a paste, which was dried in vacuum at 95°C. P-ethoxyaniline is heated to 70°C, then the above product is added, the temperature is raised to 100°C, and the reaction is performed for 3-6h to the end. Dilute with diluted hydrochloric acid at 70°C, filter, wash with hot water, and dry to obtain the product.

Uses

It is used for dyeing wool, silk, nylon and other fabrics, and also for dyeing biochemical research on leather and feathers. Used to detect protein bands after electrophoresis. It is 5-10 times more sensitive than Naphthol Blue Black B and Bromophenol (can detect 0.5μg/cm2 protein on the gel matrix). Triphenylmethane dye used for protein analysis in SDS-PAGE can also be widely used in Bradford protein quantitative reaction.


Acid Blue 83 is a dye commonly used in laboratories or quantify proteins.


Brilliant Blue R is used for detection of protein bands following electrophoresis. It is a sensitive stain for protein detection in polyacrylamide gels, typically giving blue bands on a clear background with a sensitivity of 50-100 ng/band. It may be combined with other stains, such as silver stain, to distinguish different types of proteins. It displays metachromasia by appearing pink-red, rather than blue, when binding certain proteins, such as collagen and histone H1.
