Description
Gallein is a G protein βγ (Gβγ) subunit signalling inhibitor. Gallein disrupts the interaction of Gβγ subunits with the PI3Kγ. Anticancer agent[1]. Gallein used as a red dye, an acid-base indicator and as a detection reagent for phosphates[2].
Gallein is a xanthene dye that is fluoran carrying four hydroxy substituents at positions 3', 4', 5' and 6'. It has a role as a fluorochrome, a histological dye and a G-protein-coupled receptor antagonist. It is a gamma-lactone, a polyphenol, a xanthene dye, an oxaspiro compound, an organic heteropentacyclic compound and a member of 2-benzofurans. It derives from a fluoran.

Gallein Use and Manufacturing
Derived from the reaction of pyrogallol and phthalic anhydride. Slowly heat 148g of phthalic anhydride and 126g of pyrogallol. After liquefaction, continue to heat up to 190-200°C under stirring. After 4-5h, the reactant becomes thicker and thicker until it cannot be stirred. After cooling, wash the product with hot water, add 10% ammonium hydroxide solution to dissolve, and filter out insoluble matter. The filtrate was acidified with hydrochloric acid and a brown precipitate precipitated. Filter and wash with warm water. Recrystallize with ammonia water once to obtain about 100g of finished product.
Used as acid-base indicator, pH 3.8 (brown yellow)-6.6 (rose red); used as complex indicator, titrate bismuth and zirconium, and determine phosphate in urine. This product is bright red with tetravalent tin and is a sensitive reagent for determining tin.
