Mecarbam is an organic thiophosphate that is O,O-diethyl hydrogen phosphorodithioate in which the hydrogen attached to a sulfur is replaced by a 2-[(ethoxycarbonyl)(methyl)amino]-2-oxoethyl group. It has a role as an EC 3.1.1.7 (acetylcholinesterase) inhibitor, an acaricide and an agrochemical. It is an organic thiophosphate, an organothiophosphate insecticide and a carbamate ester.

Toxicity

Mecarbam's possible production and use as an insecticide and acaricide(1) may result in its direct release to the environment through various waste streams(SRC).

AT RECOMMENDED RATES OF USE IT PERSISTS IN SOIL FOR 4-6 WEEKS.|TERRESTRIAL FATE: Based on a recommended classification scheme(1), an estimated Koc value of 420(SRC), determined from an estimated log Kow(2,SRC) and a recommended regression-derived equation(3), suggests that mecarbam will have moderate mobility in soil(SRC). Mecarbam added to a loam soil in a closed system had a half-life of 77 days(4); when added to field sites, an average half-life of 13 days was reported during June-July(4). Degradation was considerably slower from August through October(4). Both biodegradation and hydrolysis are expected to contribute to mecarbam's degradation in soil(SRC). Mecarbam may hydrolyze slowly in moist soil; a half-life of 5.9 hours was measured for mecarbam at 70 °C in an ethanol-pH 6.0 buffer solution (20:80)(5). At 20 °C this rate should be several hundred times slower(5). Volatilization of mecarbam should not be important from moist soil surfaces(SRC) given an estimated Henry's Law constant of 9.0X10-10 atm-cu m/mole(6,SRC), but may be important from dry soil surfaces(SRC) based on an estimated vapor pressure of 1.3 mm Hg(7,SRC).|AQUATIC FATE: If released to water, mecarbam may hydrolyze slowly; a half-life of 5.9 hours was measured for this compound at 70 °C in an ethanol-pH 6.0 buffer solution (20:80)(1). At 20 °C this rate should be several hundred times slower(1). Based on field studies in soil, biodegradation may also be an important fate process. In field studies, this compound had a half-life of 13 days in soil(2). Based on a recommended classification scheme(3), an estimated Koc value of 420(SRC), determined from an estimated log Kow(4,SRC) and a recommended regression-derived equation(5), indicates that mecarbam should not adsorb to suspended solids and sediment(SRC) in the water. Mecarbam will not volatilize from water surfaces based on an estimated Henry's Law constant of 9.0X10-10 atm-cu m/mole(SRC), developed using a fragment constant estimation method(6). According to a classification scheme(7), an estimated BCF value of 32(5,SRC), from an estimated log Kow(4,SRC), suggests that bioconcentration in aquatic organisms is moderate, not high(SRC) .|ATMOSPHERIC FATE: According to a suggested classification scheme(1), an estimated vapor pressure of 1.3 mm Hg at 25 °C(2,SRC) indicates that mecarbam will exist mainly in the vapor phase in the ambient atmosphere. Vapor-phase mecarbam is degraded in the atmosphere by reaction with photochemically produced hydroxyl radicals(SRC); the half-life for this reaction in air is estimated to be about 3 hours(3,SRC).|COMPARISON OF DISAPPEARANCE OF 8 ORGANOPHOSPHORUS INSECTICIDES FROM SOIL IN LAB & IN OUTDOOR EXPERIMENTS USING GAS CHROMATOGRAPHY: MECARBAM T/2 LAB= 77 DAYS; T/2 OUTDOOR= 13 DAYS. MECARBAM WAS OF INTERMEDIATE PERSISTENCE AMONG THOSE TESTED.

The rate constant for the vapor-phase reaction of mecarbam with photochemically produced hydroxyl radicals has been estimated as 1.2X10-10 cu cm/molecule-sec at 25 °C(SRC) using a structure estimation method(1,SRC). This corresponds to an atmospheric half-life of about 3 hours at an atmospheric concentration of 5X10+5 hydroxyl radicals per cu cm(1,SRC). A half-life of 5.9 hours was measured for mecarbam at 70 °C in an ethanol-pH 6.0 buffer solution (20:80)(2)(ethanol will act to slow the half-life)(3). At 20 °C this rate should be several hundred times slower(2). In a field experiment, mecarbam sprayed on apple trees had a half-life of 7.6 days(4). The process which attributed to this decay was a rate-determining irradiation process, apparently catalyzing oxidation; hydrolysis acts in a secondary manner(4). Mecarbam sprayed onto alfalfa crops had a half-life of 1.1 days(5).

An estimated BCF value of 32 was calculated for mecarbam(SRC), using an estimated log Kow of 2.29(1,SRC) and a recommended regression-derived equation(2). According to a classification scheme(3), this BCF value suggests that bioconcentration in aquatic organisms is moderate, not high(SRC).

The Koc of mecarbam is estimated as approximately 420(SRC), using an estimated log Kow of 2.29(1,SRC) and a regression-derived equation(2,SRC). According to a recommended classification scheme(3), this estimated Koc value suggests that mecarbam has moderate mobility in soil(SRC).

The Henry's Law constant for mecarbam is estimated as 9.0X10-10 atm-cu m/mole(SRC) using a fragment constant estimation method(1). This value indicates that mecarbam will be essentially nonvolatile from water surfaces(2,SRC). Mecarbam's estimated vapor pressure, 1.3 mm Hg(3,SRC) indicates that volatilization from dry soil may occur(SRC); volatilization from moist soil is not expected(SRC) based on its Henry's Law constant(1).

Farm products from the Region of Murcia were tested for the presence of 20 organophosphorus pesticides; the calculated daily ingestion of mecarbam in 1985-1986 was calculated as 2290 ng/person, mainly from citrus fruits(1). Mecarbam was detected at unreported concentrations and sources during a study of 27,065 samples of foods collected in the USA during 1988-1989(2). During the 1989(3) and 1990(4) FDA pesticide residue monitoring program, mecarbam was detected at unreported concentrations.

Drug Information

Following single oral doses of (14)C mecarbam to rats, tissue levels were highest in the liver and kidneys; some label was still found in the stomach up to 48 hr after administration. In rats given radiolabeled mecarbam iv, residues declined most rapidly during the first 40 min after administration; removal from the blood was nearly complete in 6 hr. Rats given single oral doses of radiolabeled mecarbam eliminated about 80-92% of the dose in the urine, 2-3% in the feces, and 0.2-5% as exhaled (14)CO2 within 48 hr. Urinary radioactivity was associated mainly with seven major polar metabolites; little or no unchanged mecarbam was found in the urine. Mecarbam in the diet of cows for days at 10 ppm was excreted in the milk at levels not exceeding 10 ppm.|- A suicidal poisoning committed by a 19-year-old pregnant woman, who ingested an unknown quantity of an aqueous solution mecarbam (trade name includes Murfotox) containing 68 g/l of S-(N-ethoxycarbonyl-N-methylcarbamoyl methyl) O,O-diethyl phosphorodithioate (CR Worthing (Ed), The Pesticide Manual, 6th end, BCPC, 1980) is described. The pesticide crossed the placental Barrier and caused the death of the fetus. Toxicologic analysis of mecarbam in several tissues and fluids of mother-fetus revealed its distribution. The analysis was performed by thin-layer chromatography and gas-1iquid chromatography. The case described serves to point out the ease with which mecarbam crossed the placental barrier, its high concentration in the lungs and the considerably higher concentrations in fetal than in mother's tissues.

Rats metabolized mecarbam primarily by hydrolysis, oxidative desulfuration and degradation of the carbamoyl moiety; O-deethylation was a minor pathway. At least 10 nonconjugated metabolites and 1 conjugate were found in the urine, but little or no unchanged mecarbam. Mecarbam orally administered to a lactating goat was metabolized mainly at the carbamoyl moiety, with minor O-deethylation; at least 6 nonconjugated metabolites appeared in the urine.
