4-PYRIDIN-3-YL-BENZOICACID Use and Manufacturing

4-(pyridin-3-yl)benzoic acid; To a solution of 4-boronobenzoic acid (1.66 g, 10 mmol) and 3-bromopyridine (1.72 g, 11 mmol) in acetonitrile (40 mL) and water (40 mL), potassium carbonate (5.5 g, 40 mmol), bis(triphenylphosphine)palladium(II) chloride (400 mg, 0.37 mmol) were added. The mixture was degassed and purged withed nitrogen. The mixture was stirred under reflux for 24 h. Then the hot suspension was filtered and concentrated to half of the original volume and washed with dichloromethane. The aquatic phase was adjusted to pH=3 with hydrochloric acid (1 M) and filtrated, washed with water. The residue was dried in vacuum to obtain 1.7 g of white solid of 4-(pyridin-3-yl)benzoic acid. Yield: 85percent. LC-MS (ESI) m/z: 200 (M+1)648mg of 4-Pyridin-3-yl-benzoic acid methyl ester (3.04mmol) is dissolved in 15ml of MeOH and 10ml of IN NaOH is added. The reaction mixture is , , , stirred at 50°C for 30min and room temperature for overnight. MeOH is removed in vacuo and the residue is acidified to pH=4.0 by IN HCI. Crystals are filtered and washed by water and dried to give 578mg of 4-pyridin-3-yl-benzoic acid (96percent). 1H- NMR (DMSO): 13.1(br, lH), 8.98 (m, 1H), 8.64(m, 1H), 8.17 (m, 1H), 8.07 (d, 2H, J=8.0Hz), 7.89(d, 2H, J=8.0Hz), 7.57 (m, 1H).Intermediate Preparation 64-Pyridin-3-yl-benzoic acid Procedure J': 648 mg of 4-Pyridin-3-yl-benzoic acid methyl ester (3.04 mmol) is dissolved in 15 ml of MeOH and 10 ml of 1N NaOH is added. The reaction mixture is stirred at 50° C. for 30 min and room temperature for overnight. MeOH is removed in vacuo and the residue is acidified to pH=4.0 by 1N HCl.
