Description
White, granular, crystalline substance; camphor-like odor. Soluble in water; freely soluble in alcohol, chloroform, and ether; pH 6.0 (5% solution).

Trimethadione is an oxazolidinone. It has a role as a geroprotector and an anticonvulsant.|An anticonvulsant effective in absence seizures, but generally reserved for refractory cases because of its toxicity. (From AMA Drug Evaluations Annual, 1994, p378)|Trimethadione is an Anti-epileptic Agent. The physiologic effect of trimethadione is by means of Decreased Central Nervous System Disorganized Electrical Activity.|Trimethadione is a dione-type anticonvulsant with antiepileptic activity. Trimethadione reduces T-type calcium currents in thalamic neurons, thereby stabilizing neuronal membranes, raising the threshold for repetitive activities in the thalamus and inhibiting corticothalamic transmission. This decreases the abnormal thalamocortical rhythmicity, which is thought to underlie the paroxysmal three-cycle-per-second spike-and-wave pattern seen with absence (petit mal) seizures.

Dione anticonvulsants are used in the treatment of epilepsy. They act on the central nervous system (CNS) to reduce the number of seizures.

Thus, the abnormal thalamocortical rhythmicity, which is thought to underlie the 3-Hz spike-and-wave discharge seen on electroencephalogram(EEG) with absence seizures, is dampened.
