In a significant breakthrough, IovanceBiotherapeutics announced FDA accelerated approval for Lifileucel (brand name Amtagvi) on February 16, 2024. Lifileucel is the world's first FDA-approved TIL therapy, designed to treat advanced melanoma that has progressed after PD-1 antibody therapy. TIL therapy involves isolating lymphocytes from tumors, expanding them in vitro, and reinfusing them into patients. With TIL cells specifically recognizing and targeting patient-specific cancer cells, this therapy offers a promising avenue for personalized cancer treatment.
2. TCR-T Therapy: Expanding Targeting to Intracellular Antigens
Adaptimmune's TCR-T therapy, Afami-cel, achieved FDA acceptance for its marketing application in the treatment of synovial sarcoma on January 31, 2024. Afami-cel holds the potential to become the first T-cell therapy for solid tumors, expanding the scope of immunotherapy to intracellular targets. Success in gaining approval would mark a significant milestone in the field, offering a systemic breakthrough for solid tumor treatment.
3. Oncolytic Viruses: Revolutionizing Treatment for Multiple Cancers
Replimune aims to submit a marketing application for its oncolytic virus therapy, RP1, for the treatment of PD-1-resistant melanoma in the second half of 2024. RP1 has demonstrated potent therapeutic potential in PD-1-resistant patients, with FDA recognition based on phase 1/2 clinical data as supporting evidence for its unmet clinical need. Replimune is also exploring the therapeutic potential of oncolytic virus therapy for various skin cancers, including CSCC. Moderna's collaboration with Merck in developing mRNA therapy targeting tumor-specific neoantigens has shown promising results in phase 2 clinical trials.
