$1 Billion Deal Between Novo Nordisk and Lexicon Pharmaceuticals for Obesity Drug

On March 28, Lexicon Pharmaceuticals announced a significant exclusive licensing agreement with Novo Nordisk for the development of LX9851, the first oral non-incretin-based candidate drug aimed at treating obesity and related metabolic disorders. The total value of this deal is estimated to be around $1 billion.


Lexicon will receive an upfront payment of up to $75 million, along with milestone payments tied to development, regulatory approvals, and sales, as well as a tiered royalty on net sales of LX9851. Under the terms of the agreement, Novo Nordisk will hold exclusive global rights for the development, production, and commercialization of LX9851.


LX9851 is a potent, selective oral small molecule inhibitor that targets Acyl-CoA Synthetase 5 (ACSL5), a key player in regulating fat accumulation and energy balance. This drug may enhance satiety by activating the ileal brake mechanism, potentially delaying gastric emptying and suppressing appetite.


Preclinical efficacy data presented during the 2024 Obesity Week indicated that LX9851, when used in conjunction with semaglutide, significantly reduced body weight, food intake, and fat mass—superior to semaglutide alone. Additionally, introducing LX9851 after discontinuing semaglutide may mitigate weight rebound and positively affect liver steatosis.


Mike Exton, CEO of Lexicon, expressed enthusiasm for the partnership, stating, "We are thrilled that Novo Nordisk recognizes the potential of LX9851. This collaboration significantly strengthens our financial position, providing us with options to further invest and accelerate our product development pipeline."


Jacob Sten Petersen, Senior Vice President at Novo Nordisk for diabetes, obesity, and MASH treatments, emphasized the company's commitment to addressing diverse patient needs in obesity and metabolic diseases. He remarked, "We are excited to explore a new biological mechanism and potential treatment paradigm with this agreement, building upon Lexicon's existing work on LX9851."
