Allopurinol Tablets
Function and Efficacy
This product is a drug that inhibits the synthesis of uric acid. Allopurinol and its metabolite oxypurinol can inhibit xanthine oxidase, prevent hypoxanthine and xanthine from being metabolized into uric acid, and thus reduce the production of uric acid. It reduces the uric acid content in blood and urine to a level below the solubility, prevents uric acid from forming crystals and depositing in joints and other tissues, and also helps to redissolve uric acid crystals in the tissues of gout patients. Allopurinol also inhibits the synthesis of new purines in the body by acting on hypoxanthine-guanine phosphate nucleic acid transferase. The blood uric acid concentration of this product begins to decrease 24 hours after oral administration, and the decrease is most obvious in 2 to 4 weeks. Acute toxicity test results: The oral LD50 of rats is 6000 mg/kg, and the intraperitoneal injection LD50 is 750 mg/kg; the oral LD50 of mice is 700 mg/kg, and the intraperitoneal injection LD50 is 160 mg/kg.
Ingredients

Appearance
This product is white or off-white tablets.
Indication
1. Primary and secondary hyperuricemia, especially hyperuricemia caused by excessive uric acid production; 2. Patients with recurrent or chronic gout; 3. Tophi; 4. Uric acid kidney stones and/or uric acid nephropathy; 5. Hyperuricemia with renal insufficiency.
Usage and Dosage
Common oral dosage for adults: initial dose 50 mg once, 1 to 2 times a day, can be increased by 50 to 100 mg per week to 200 to 300 mg per day, divided into 2 to 3 doses. Measure blood and urine uric acid levels every 2 weeks. If they have reached normal levels, no more increments will be made. If they are still high, they can be increased again. However, the maximum daily dose should not exceed 600 mg. 2 Common dosage for children to treat secondary hyperuricemia: 50 mg each time for children under 6 years old, 1 to 3 times a day; 100 mg each time for children aged 6 to 10 years old, 1 to 3 times a day. The dosage can be adjusted as appropriate.
Adverse Reactions
(1) Rash: It may be pruritic papules or urticaria. If the rash is widespread and persistent, symptomatic treatment is ineffective, and there is a tendency to worsen, the drug must be discontinued. (2) Gastrointestinal reactions: including diarrhea, nausea, vomiting and abdominal pain. (3) If leukopenia, thrombocytopenia, anemia, or bone marrow suppression occur, drug discontinuation should be considered. (4) Other symptoms include hair loss, fever, lymphadenopathy, hepatotoxicity, interstitial nephritis and allergic vasculitis. (5) Several cases of sudden death of patients with unknown causes while taking this product have been reported abroad.
Precautions
It is contraindicated for patients who are allergic to this product, have severe liver and kidney dysfunction, or have obvious low blood cells.
Special Population Medication
Precautions for children: The dosage for children should be adjusted as appropriate. Precautions for pregnancy and lactation: Forbidden. Precautions for the elderly: The elderly should use the drug with caution and reduce the daily dosage.
Drug Interactions
1 Drinking alcohol, diuretics such as chlorthalidone, uric acid, furosemide, pyrazinamide and thiazide can increase serum uric acid concentration. When this product is used with these drugs, it is necessary to adjust the dosage to control hyperuricemia. 2 When used with aspirin or other salicylates, the uric acid excretion effect of this product can be inhibited. 3 When used with indomethacin, dapsone, naproxen, etc., the blood concentration of the latter increases, and the toxicity is increased. 4 When used with various penicillins and cephalosporins, the blood concentration of the latter increases and lasts for a long time, and the toxicity is increased, especially the toxicity to the kidneys. 5 When used with oral hypoglycemic drugs, the effect of the latter is enhanced.
