Ticlopidine Hydrochloride Capsules
Function and Efficacy
Ticlopidine is a platelet aggregation inhibitor. Platelet activation is affected by many factors, among which adenosine diphosphate (ADP) plays a key role. When adenosine diphosphate binds to its specific receptor, it can activate the fibrinogen receptor (glycoprotein IIb-IIIa complex) on the surface of the platelet membrane, and bind to fibrinogen to cause platelet aggregation (phase I aggregation). In addition, after platelet activation, adenosine diphosphate can be released, leading to further platelet aggregation (phase II aggregation). Ticlopidine has a strong inhibitory effect on ADP-induced platelet aggregation (including phase I and phase II aggregation), and the effect is long-lasting. In addition, ticlopidine can reduce fibrinogen concentration and blood viscosity, and increase the filtration rate of whole blood and red blood cells.

Indication
Prevent and treat circulatory disorders of the heart, brain and other arteries caused by high platelet aggregation.
Usage and Dosage
Oral administration: 1 tablet (0.25 g) once a day; take with meals to reduce gastrointestinal reactions.
Adverse Reactions
1. Occasionally mild gastrointestinal reactions; 2. Rare reactions include: nausea, diarrhea, rash, ecchymosis, gingival bleeding, leukopenia, granulocytopenia, bile obstructive jaundice, liver damage. The above adverse reactions disappear after discontinuation of the drug.
Precautions
1. Patients with hemophilia or other bleeding diseases, patients with granulocytopenia or thrombocytopenia, patients with ulcer disease and patients with active bleeding should not use this drug; 2. Patients with severe liver damage often increase the risk of bleeding due to coagulation factor synthesis disorders, so this product should not be used; 3. People who are allergic to this product.
Drug Interactions
1. The combination of this product with any platelet aggregation inhibitor, thrombolytic agent and drug that causes hypoprothrombinemia or thrombocytopenia can increase the risk of bleeding. If it is clinically necessary to use the drug together, close observation and laboratory monitoring should be performed; 2. When this product is used in combination with theophylline, theophylline blood concentration will increase and there is a risk of overdose because it reduces the clearance rate of the latter. Therefore, the dosage of theophylline should be adjusted during and after the use of this product, and theophylline blood concentration should be monitored when necessary; 3. When this product is used in combination with digoxin, the latter's blood concentration can be slightly reduced (about 15%), but it generally does not affect the clinical efficacy of digoxin; 4. Occasionally, there are reports that this product reduces the blood concentration of cyclosporine, so the blood concentration of cyclosporine should be monitored regularly when the two are used in combination.
