J147 is a very effective and oral neurotrophic molecule with an EC 50 of about 25 nm. In rat hippocampal neurons, j147 can resist the toxicity of amyloid protein, and its EC50 is about 200 nm. In transgenic mice with Alzheimer's disease, j147 can prevent synaptic protein loss and cognitive decline. Increased levels of brain-derived neurotrophic factor (BDNF) and expression of BDNF related proteins, enhanced LTP, preservation of synaptic proteins, decreased markers of oxidative stress and inflammation, decreased amyloid plaques, and lower levels of soluble a β 1-42 and a β 1-40 are related to the neurotrophic effect and memory enhancing activity of j147. In the old mouse model of Alzheimer's disease, the memory promoting function of j147 is also related to the neurotrophic factor NGF induced by j147. These multiple effects of a single molecule indicate that j147 has potential for the treatment of Alzheimer's disease (AD).

J-147 was discovered based upon efficacy in multiple cell culture models of age-associated pathologies rather than exclusively amyloid metabolism. It is an exceptionally potent, orally active and broadly neuroprotective compound has the ability to enhance memory in normal animals as well as to prevent memory deficits in Alzheimer's disease (AD) transgenic mice. The neurotrophic and memory-enhancing activities of J147 are associated with an increase in brain derived neurotrophic factor (BDNF) levels and the expression of BDNF responsive proteins, the enhancement of LTP, the preservation of synaptic protein, the reduction of amyloid plaques, etc.
