    Description


    Heptakis(2,6-di-O-methyl)-β-cyclodextrin is a cholesterol sequestering agent which modifies lipid rafts of the cell membrane. β-cyclodextrins are involved in cytotoxicity in various cells through complexation with cholesterol and membrane lipids.

    Ionic forms of these molecules greatly reduce its cytoxicity compared to its methylated form. The cytotoxicity of β-cyclodextrins is affected by the position of the substitution of the methylated portion of the molecule.

    The ability of a β-cyclodextrin to potentiate cytotoxic effects is dependent on the molecules ability to extract cholesterol from cell membranes. Alternate studies indicate that 2, 6-Di-O-Methyl-Beta-Cyclodextrin contains a stereoselective cavity only when the R substituent is linked to the chiral carbon which contains an aromatic ring.

    Biological Activity

    Methyl-β-cyclodextrin is a cyclodextrin derivative that can form inclusion complexes with cholesterol molecules. By sequestering cholesterol, it disrupts the integrity of lipid rafts, which are specialized microdomains in the cell membrane involved in various cellular processes, including signal transduction.

    Studies have shown that Methyl-β-cyclodextrin treatment induces caspase-dependent apoptosis in PEL cells. Caspases are a family of proteases involved in programmed cell death. The induction of apoptosis by Methyl-β-cyclodextrin leads to the inhibition of PEL cell growth and invasion.
