Buvacirtan was approved by the US Food and Drug Administration (FDA) on February 19th, mainly for the adjuvant treatment of partial epileptic seizures in patients aged 16 years and above. Compared with the previous-generation drug levetiracetam, buvasirtan has a similar chemical structure and mechanism of action. It is the first antiepileptic drug approved by the FDA for the treatment of partial epileptic seizures since 2013. Indication: brivaracetam is a highly selective and affinity ligand of synaptic vesicular protein 2A, approved for the adjuvant treatment of partial seizures in adult and adolescent epilepsy patients over 16 years old, with or without secondary generalized seizures. The main function of buvacicetan is that it has a high Chemicalbook affinity and can selectively bind to synaptic vesicle protein 2A(SV2A), which is the action site of levetiracetam in AED. SV2A is located on the presynaptic membrane and participates in regulating the release of neurotransmitters and vesicle circulation, thereby maintaining the normal function of synaptic vesicles. The combination of AED and SV2A can reduce the release of excitatory neurotransmitters and control epileptic seizures by regulating the balance of excitatory and inhibitory neurotransmitters in the brain. The affinity of Bovacirtan is 15 to 30 times that of levetiracetam, reducing its dosage by approximately 10 times. Buvasicitan, as a new generation of antiepileptic drugs, has been approved by the FDA for the treatment of partial epileptic seizures in epilepsy patients aged 16 years and above. It can be used in laboratory research and the pharmaceutical development process.
