Osimertinib, as a third-generation EGFR inhibitor, is targeted at the patient subgroup with EGFRT790M mutations, meeting such unmet market demands: Osimertinib has high selectivity. The IC50 scores for T790M/L858R mutation and exon 19 deletion EGFR are 11.4nM and 12.9nM respectively, while the inhibitor activity (IC50) for wild-type EGFR is 494nM. Osimertinib is an irreversible inhibitor of EGFR and can form covalent bonds with molecular targets. Therefore, theoretically, the response lasts longer and the chance of developing drug resistance is correspondingly reduced.
