It serves as a precursor to nicotinamide adenine dinucleotide (NAD+), a coenzyme critical for cellular metabolism. As an active pharmaceutical ingredient (API), nicotinamide is commonly formulated in tablets, capsules, and injectable solutions for medical and nutritional applications.

Mechanism of Action

Nicotinamide functions primarily by converting into NAD+ and NADP+, which are vital for redox reactions, DNA repair, and energy production in cells. It inhibits poly(ADP-ribose) polymerase (PARP) and modulates inflammatory pathways, making it beneficial in treating conditions like pellagra, diabetes-related skin disorders, and certain autoimmune diseases. Unlike niacin, it does not cause vasodilation (flushing).

General supplementation : 50–500 mg daily, depending on deficiency or therapeutic need.

Pellagra treatment : 300–500 mg/day in divided doses.

Topical use (dermatology) : 2–5% concentration in creams.
